肺部惡性腫瘤原代裸鼠移植瘤模型的建立

【摘要】:背景與目的動物模型是研究癌癥的重要工具。本研究的目的是建立人肺部惡性腫瘤組織裸鼠移植瘤的成熟方法,并積累一定數(shù)量的動物模型,為肺部惡性腫瘤提供有效的研究平臺。 方法將手術切除的肺部惡性腫瘤組織的新鮮標本移植于裸鼠皮下,觀察移植瘤的生長情況,待皮下移植瘤長成后對其進行連續(xù)傳代和組織學觀察。選用裸鼠體內(nèi)傳代的移植瘤模型進行體內(nèi)藥效實驗。 結果共移植101例人肺部惡性腫瘤的組織標本,其中93例術后病理證實為原發(fā)性非小細胞肺癌,3個月內(nèi)成瘤27例(29%);3例為肺轉(zhuǎn)移癌,成瘤率為100%。各代移植瘤模型的組織病理學與人源腫瘤組織相一致。對1例原發(fā)于子宮內(nèi)膜的肺轉(zhuǎn)移癌裸鼠移植瘤模型進行了體內(nèi)藥效試驗,結果顯示紫杉醇、順鉑和吉西他濱能在不同程度上抑制裸鼠移植瘤的生長。 結論成功建立了肺部惡性腫瘤的裸鼠皮下移植瘤模型,并較好地保留了人源腫瘤的惡性特征和組織學特性,可為肺部惡性腫瘤的研究提供良好的實驗平臺。
[Abstract]:Background and objective Animal models are important tools for cancer research. The purpose of this study was to establish a mature method for transplanted human lung malignant tumors in nude mice and to accumulate a certain number of animal models to provide an effective platform for the study of lung malignant tumors. Methods fresh specimens of lung malignant tumor were transplanted into nude mice subcutaneously, the growth of transplanted tumor was observed, and the subcutaneous transplanted tumor was subcultured and histologically observed. The model of xenograft tumor in nude mice was used to carry out in vivo pharmacodynamics experiment. Results among 101 cases of lung cancer, 93 cases were confirmed as primary non-small cell lung cancer (NSCLC) after operation, 27 cases (29%) were transplanted within 3 months, and 3 cases were lung metastases, the rate of tumorigenesis was 100 cases. The histopathology of various generations of transplanted tumor models is consistent with that of human tumor tissues. A nude mouse model of lung metastatic carcinoma derived from endometrium was tested in vivo. The results showed that paclitaxel, cisplatin and gemcitabine could inhibit the growth of transplanted tumor in nude mice to some extent. Conclusion the subcutaneous transplanted tumor model of lung malignant tumor was successfully established in nude mice, and the malignant and histological characteristics of human tumor were well preserved, which could provide a good experimental platform for the study of lung malignant tumor.
