【摘要】：目的構(gòu)建具有能同時阻斷白細胞介素12(IL-12)/IL-23 p40與腫瘤壞死因子α(TNF-α)的雙功能抗體,并對其生物學(xué)功能進行鑒定,觀察其對小鼠銀屑病的阻斷效果。方法根據(jù)已有的阿達木單抗(adalimumab)和優(yōu)特克單抗(ustekinumab)蛋白序列,設(shè)計、構(gòu)建并制備了全新的雙功能抗體Bi AU003、Bi AU022和Bi AU023。采用ELISA檢測抗原抗體結(jié)合能力;用TNF-α和雙功能抗體處理人臍靜脈內(nèi)皮細胞(HUVEC),加入FITC標(biāo)記的內(nèi)皮白細胞黏附分子1(ELAM-1)抗體,經(jīng)流式細胞術(shù)檢測ELAM-1的表達量;用IL-12和雙功能抗體處理經(jīng)IL-2活化的人外周血單個核細胞(PBMC),并用ELISA檢測上清液中γ干擾素(IFN-γ)的量;體外實驗采用雙功能抗體治療IL-12和TNF-α誘導(dǎo)的銀屑病小鼠,HE染色檢測病變皮膚厚度。結(jié)果3種雙功能抗體對其相應(yīng)的抗原都有較強的親和力,能明顯抑制ELAM-1蛋白的表達和IFN-γ的分泌,并且明顯抑制小鼠的銀屑病形成,其效果與現(xiàn)有的抗體藥阿達木單抗和優(yōu)特克單抗相當(dāng)或更優(yōu)。結(jié)論新構(gòu)建的3種雙功能抗體Bi AU003、Bi AU022和Bi AU023,是TNF-α和IL-12/IL-23 p40的阻斷劑,能有效阻斷小鼠銀屑病形成。
[Abstract]:Objective to construct a bifunctional antibody with simultaneous blocking of interleukin 12 (IL-12) / IL-23 p40 and tumor necrosis factor 偽 (TNF- 偽), and to identify its biological function and to observe its blocking effect on psoriasis in mice. Methods A novel bifunctional antibody, Bi AU003,Bi AU022 and Bi AU023., was designed, constructed and prepared according to the existing Adamumab (adalimumab) and UTC McAb (ustekinumab) protein sequences. Antigen-antibody binding ability was detected by ELISA. Human umbilical vein endothelial cells (HUVEC),) were treated with TNF- 偽 and bifunctional antibodies to FITC labeled endothelial leukocyte adhesion molecule-1 (ELAM-1) antibody. The expression of ELAM-1 was detected by flow cytometry. Human peripheral blood mononuclear cells (PBMC),) activated by IL-2 were treated with IL-12 and bifunctional antibody, and the amount of interferon 緯 (IFN- 緯) in supernatant was detected by ELISA. In vitro, bifunctional antibodies were used to treat psoriasis induced by IL-12 and TNF- 偽. The thickness of diseased skin was detected by HE staining. Results all of the three bifunctional antibodies had strong affinity to the corresponding antigens, and could significantly inhibit the expression of ELAM-1 protein and the secretion of IFN- 緯, as well as the formation of psoriasis in mice. The effect is comparable to or better than that of the existing antibodies adamumab and eutectoma. Conclusion three newly constructed bifunctional antibodies, Bi AU003,Bi AU022 and Bi AU023, are antagonists of TNF- 偽 and IL-12/IL-23 p40, which can effectively block the formation of psoriasis in mice.
