結(jié)核分枝桿菌Rv2986c對小鼠樹突狀細胞成熟和功能的影響

【摘要】:目的探索結(jié)核分枝桿菌Rv2986c蛋白誘導小鼠樹突狀細胞成熟及Na?ve CD4~+T細胞極化的作用。方法克隆表達Rv2986c蛋白,用純化后的蛋白刺激小鼠樹突狀細胞,流式檢測樹突狀細胞MHC-Ⅱ表面分子,ELISA檢測培養(yǎng)上清中IL-6和IL-12p40分泌情況。將Na?ve CD4~+T細胞與經(jīng)Rv2986c刺激后的樹突狀細胞共培養(yǎng),檢測培養(yǎng)上清中IFN-γ的分泌水平。結(jié)果成功克隆表達了Rv2986c蛋白。經(jīng)Rv2986c蛋白刺激后,樹突狀細胞MHC-Ⅱ表面分子表達水平顯著增高,IL-6和IL-12p40釋放水平顯著升高。淋巴細胞共培養(yǎng)上清液中IFN-γ分泌量顯著增加。結(jié)論結(jié)核分枝桿菌蛋白Rv2986c能促進小鼠樹突狀細胞成熟,激活抗原遞呈作用,誘導CD4~+T向Th1型極化。
