【摘要】：目的：觀察肺、腎、食管、胃、小腸、大腸在人胎兒發(fā)育過程中的形態(tài)學變化,檢測巢蛋白在人胎兒不同時期肺、腎、食管、胃、小腸、大腸組織的表達及其變化,并推測巢蛋白在人胎兒發(fā)育過程中的作用。 方法：取3-8月人胎兒肺、腎、食管、胃、小腸、大腸,常規(guī)石蠟切片并將其分為兩組,一組作HE染色觀察這些組織在發(fā)育過程中的形態(tài)學變化,一組作免疫組織化學染色,檢測巢蛋白在胎兒發(fā)育過程這些組織中的表達,根據(jù)免疫組化結(jié)果進行半定量分析以判斷巢蛋白在不同時期胎兒組織的表達變化。 結(jié)果：巢蛋白在人胎兒發(fā)育各個時期的肺、腎、食管、胃、小腸、大腸組織均有表達。巢蛋白在肺組織主要表達在支氣管旁、肺間質(zhì)和微血管以及毛細血管壁上,而在肺導氣部管道黏膜上皮沒有陽性表達。巢蛋白在腎組織主要表達在腎小球、腎小囊壁層和微血管及毛細血管壁上,在近端小管、遠端小管的上皮中沒有表達。巢蛋白在消化管主要表達在固有層、黏膜下層和肌層內(nèi)的微血管和毛細血管壁上巢蛋白在這些組織的表達均隨胎齡的增加而減少。 結(jié)論：巢蛋白在胎兒發(fā)育過程的多種組織中均有表達,且表達強度隨胎齡的變化而變化,提示巢蛋白可能與胎兒發(fā)育有關(guān)。另外,巢蛋白在胎兒多種組織的的微血管和毛細血管的管壁上均有表達,且隨著胎兒發(fā)育,其表達逐漸減少,提示巢蛋白在這些器官組織的血管形成過程中起作用。
[Abstract]:Objective: to observe the morphological changes of lung, kidney, esophagus, stomach, small intestine and large intestine during the development of human fetus, and to detect the expression and changes of nestin in lung, kidney, esophagus, stomach, small intestine and large intestine of human fetus at different stages. The role of nestin in the development of human fetus was speculated. Methods: the human fetal lung, kidney, esophagus, stomach, small intestine, large intestine and normal paraffin sections were divided into two groups: one group was stained with HE to observe the morphological changes of these tissues during development, the other group was stained with immunohistochemistry. The expression of nestin in these tissues during fetal development was detected and semi-quantitative analysis was carried out according to the immunohistochemical results to determine the changes of nestin expression in fetal tissues at different stages. Results: nestin was expressed in lung, kidney, esophagus, stomach, small intestine and large intestine of human fetus. The expression of nestin in lung tissue was mainly near the bronchus, pulmonary interstitial and microvessel and capillary wall, but not in the mucosal epithelium of pulmonary duct. Nestin was mainly expressed in glomeruli, renal capsule wall, microvessel and capillary wall, but not in proximal tubule and distal tubule epithelium. Nestin was mainly expressed in the lamina propria in the digestive tract. The expression of nestin in the wall of microvessels and capillaries in the submucosa and myometrium decreased with the increase of gestational age. Conclusion: nestin is expressed in various tissues during fetal development, and the expression intensity varies with gestational age, suggesting that nestin may be related to fetal development. In addition, nestin was expressed on the walls of microvessels and capillaries in many kinds of fetal tissues, and decreased with the development of fetus, suggesting that nestin plays an important role in the angiogenesis of these organs.
