【摘要】：目的：本實(shí)驗(yàn)旨在中醫(yī)理論指導(dǎo)下,探討復(fù)制宮環(huán)出血病陰虛血瘀證動物模型的方法,并對其發(fā)病機(jī)理進(jìn)行研究。 方法：本文回顧了中西醫(yī)關(guān)于陰虛血瘀型宮環(huán)出血病的研究現(xiàn)狀和進(jìn)展,將大鼠按隨機(jī)數(shù)字表分為四組：空白組、置環(huán)組、模型組、治療組。通過手術(shù)置環(huán)結(jié)合藥物灌胃的復(fù)合手法構(gòu)建宮環(huán)出血病陰虛血瘀型大鼠模型,觀察各組大鼠一般行為改變、體重變化以及大鼠子宮形態(tài)學(xué)改變,并通過治療組對比觀察,最后利用蛋白質(zhì)抗體芯片對大鼠子宮組織中細(xì)胞因子表達(dá)水平進(jìn)行檢測,觀察各組中細(xì)胞因子變化情況,通過藥物對細(xì)胞因子的干預(yù)情況來進(jìn)一步佐證動物模型。 結(jié)果：模型組大鼠行為異常,主要表現(xiàn)為飲食飲水增加、活動度增加,興奮性提高,易激怒,毛色失去光澤,生長較對照組緩慢,其差異具有高度統(tǒng)計(jì)學(xué)意義(P0.01)。子宮組織形態(tài)涂片呈現(xiàn)內(nèi)膜上皮細(xì)胞壞死脫落,炎癥細(xì)胞浸潤等。與置環(huán)組相比,模型組中白介素-1β(IL-1β)、腫瘤細(xì)胞壞死因子-α(TNF-α)、單核細(xì)胞趨化蛋白-1(MPC-1)以及基質(zhì)金屬蛋白酶組織抑制因子(TIMP-1)4種細(xì)胞因子的表達(dá)水平發(fā)生了顯著變化。其中IL-1β、TNF-α水平同空白組相比升高,其差異具有高度統(tǒng)計(jì)學(xué)意義(P0.01),MCP-1較空白組顯著升高,其差異具有統(tǒng)計(jì)學(xué)意義(P0.05), TIMP-1較空白組顯著降低,其差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論：實(shí)驗(yàn)證明置環(huán)結(jié)合藥物灌胃的復(fù)合式造模方法比單一的置環(huán)能更顯著的影響大鼠體重、細(xì)胞形態(tài)學(xué)以及組織內(nèi)細(xì)胞因子表達(dá)水平。藥物治療佐證了模型的合理性。蛋白質(zhì)抗體芯片能平行的檢測多種細(xì)胞因子的表達(dá)水平,初步估計(jì)IL-1β、TNF-α、MPC-1以及TIMP-1四種細(xì)胞因子與陰虛血瘀有關(guān),但仍需大量實(shí)驗(yàn)證明。
[Abstract]:Objective: under the guidance of traditional Chinese medicine (TCM) theory, the aim of this experiment was to explore the method of replicating the animal model of uterine ring hemorrhage with yin deficiency and blood stasis, and to study its pathogenesis. Methods: the present situation and progress of traditional Chinese and western medicine on uterine ring haemorrhage of yin deficiency and blood stasis type were reviewed. Rats were randomly divided into four groups: blank group, ring setting group, model group and treatment group. A rat model of uterine ring hemorrhage with yin deficiency and blood stasis was established by the combined manipulation of surgical ring placement and drug administration. The changes of general behavior, body weight and uterine morphology of rats in each group were observed and compared with those in the treatment group. Finally, the expression level of cytokines in rat uterus was detected by protein antibody chip, and the changes of cytokines in each group were observed, and the animal model was further confirmed by the intervention of drugs on cytokines. Results: the rats in the model group showed abnormal behavior, mainly manifested by the increase of diet and drinking water, the increase of activity, the increase of excitability, the irritation, the loss of luster and the slower growth of the model group than those in the control group (P0.01). Uterine tissue morphologic smears showed necrosis and exfoliation of endometrial epithelial cells and infiltration of inflammatory cells. The expression levels of interleukin-1 尾 (IL-1 尾), tumor necrosis factor- 偽 (TNF- 偽), monocyte chemoattractant protein-1 (MPC-1) and tissue inhibitor of matrix metalloproteinase (TIMP-1) in the model group changed significantly compared with those in the annular group. The levels of IL-1 尾 and TNF- 偽 increased compared with the blank group, the difference was statistically significant (P0.01), MCP-1 was significantly higher than the blank group, the difference was statistically significant (P0.05), the TIMP-1 was significantly lower than the blank group, the difference was statistically significant (P0.05). Conclusion: the experimental results showed that the combined method of intragastric perfusion with ring insertion could significantly affect the body weight, cell morphology and cytokine expression level of rats. Drug therapy proved the rationality of the model. Protein antibody chip could detect the expression level of many cytokines in parallel. It was estimated that four cytokines, IL-1 尾, TNF- 偽, MPC-1 and TIMP-1, were related to blood stasis due to yin deficiency, but a large number of experiments were needed to prove them.
