大鼠虛寒證模型的建立、評價(jià)及附子對虛寒證大鼠影響的研究
[Abstract]:Objective: to explore the model and evaluation method of the model of asthenia cold syndrome in rats induced by cold drugs. By observing the effect of aconite on energy metabolism and related indexes in rats with deficiency and cold syndrome, the effect trend, characteristics and rules of aconite were analyzed, and the objective indexes reflecting the cold and heat properties of experimental animals were explored. To reveal the internal relationship between the drug properties of cold and heat, and to lay a foundation for better guidance of clinical drug use. Methods: the rat model of deficiency cold syndrome was established by oral administration of raw gypsum, gentian grass, Phellodendron chinensis and Anemarrhena chinensis for 14 days. The rats were treated with water decoction of aconite for 7 days. The changes of general condition, body temperature, cold and heat tendency of rats with deficiency cold were observed. The intake energy, digestibility energy, fecal energy and metabolizable energy of rats were measured by oxygen bomb calorimeter, and the contents of T3T4 TSHTSHTRH in plasma were detected by enzyme-linked immunosorbent assay (Elisa). Fasting blood glucose content, liver glycogen, lactic acid content, ATP enzyme activity, lactate dehydrogenase activity, (LDH), succinate dehydrogenase (SDH) activity, liver tissue ATP content and hepatocyte energy charge were detected by HPLC. Results: at the end of the model making, the model group showed a marked decrease in hair color, diarrhea, body weight, spontaneous activity and toe temperature, and the contents of plasma pyruvate and T3 decreased. Aconite could significantly increase plasma T4 content and liver lactate content, increase the activity of liver lactate dehydrogenase and liver Na~ K- ATP enzyme, and increase hepatic energy charge, fasting blood glucose, body weight, anus temperature, drinking water and urine volume. In the hot box, the number of fecal weight, the number of intake energy per unit body weight, fecal energy, succinate dehydrogenase activity and ATP content showed a certain increasing trend. Conclusion: the cold drug gypsum, gentian grass, Phellodendron chinensis and Anemarrhena can be successfully used to replicate the model of deficiency cold syndrome in rats. Radix aconite can enhance the energy metabolism and improve the cold state in rats with deficiency cold. Its pharmacological effect is closely related to the enhancement of liver energy metabolism and the promotion of thyroid hormone secretion. Body weight, body temperature, plasma thyroid hormone content, liver lactic acid content, ATP enzyme activity, liver cell energy charge and fasting blood glucose content can be used as important evaluation indexes of cold and heat syndrome model and research of cold and heat Chinese medicine.
