苯肼誘發(fā)昆明種小鼠溶血性貧血模型的建立
[Abstract]:Objective to observe the changes of mature erythrocytes and reticulocytes in peripheral blood of Kunming mice after intraperitoneal injection of phenylhydrazine and to establish hemolytic anemia model of Kunming mice. Methods 18 healthy male Kunming mice were randomly divided into three groups: solvent control group, low dose phenylhydrazine group and high dose phenylhydrazine group. Rats in the control group were intraperitoneally injected with normal saline on the 0 th day. The animals in the low dose phenylhydrazine group and the high dose phenylhydrazine group were intraperitoneally injected with phenylhydrazine for 30 and 60 mg/kg bw, respectively. After that, erythrocyte count and reticulocyte count were collected from the tail of the rat every day. Results after the injection of phenylhydrazine, the total number of erythrocytes in peripheral blood and mature erythrocytes in the low dose phenylhydrazine group and the high dose phenylhydrazine group decreased gradually, and reached the lowest on the 5th and 4th day, respectively. The decrease of high dose phenylhydrazine group was significantly higher than that of low dose phenylhydrazine group, and after that, the erythrocyte count and mature erythrocyte count began to rise gradually, and returned to the control level on the 13th day. At the same time, the total number and proportion of reticulocyte in peripheral blood of mice gradually increased from the first day after injection of phenylhydrazine, and the total and proportion of reticulocyte in low dose group reached the highest level on the 6th day. The total number of high dose reticulocyte reached the highest on the 5th day, the reticulocyte ratio reached the highest on the 4th day, then gradually fell back to normal level on the 13th day. Conclusion A single intraperitoneal injection of phenylhydrazine can induce mild hemolytic anemia in Kunming mice and stimulate hematopoietic tissue erythropoiesis in order to restore the level of peripheral blood red blood cells. The total number of red blood cells and reticulocytes in peripheral blood can reflect the progression of anemia and the hemopoietic function of the hemolytic anemia model.
