【摘要】：目的:觀察不同濃度粒-巨噬細(xì)胞集落刺激因子(granulocyte-macrophage colonystimulating factor,GM-CSF)聯(lián)合白介素4(interleukin,IL-4)對小鼠骨髓源性樹突狀細(xì)胞(dendritic cell,DC)形態(tài)、純度、細(xì)胞數(shù)目的影響,探究樹突狀細(xì)胞誘導(dǎo)培養(yǎng)的最適宜條件.方法:分離、純化小鼠骨髓單個(gè)核樣細(xì)胞,分別加入不同濃度梯度的GM-CSF和定量的IL-4,對其進(jìn)行形態(tài)學(xué)觀察和細(xì)胞表型檢測,以及對異基因淋巴細(xì)胞的激活作用測定.結(jié)果:在一定濃度范圍內(nèi),隨著GM-CSF濃度的增加,小鼠骨髓源性樹突狀細(xì)胞的數(shù)目、純度及成熟度亦隨之增加.而過高濃度的GM-CSF反而導(dǎo)致抑制作用.據(jù)此,可推測理想的GM-CSF濃度范圍應(yīng)為10-20 ng/mL.結(jié)論:適宜濃度的GM-CSF聯(lián)合IL-4可提升小鼠骨髓單個(gè)核樣細(xì)胞向樹突狀細(xì)胞分化的能力,兩種細(xì)胞刺激因子的合理運(yùn)用,可增加樹突狀細(xì)胞的數(shù)目、純度及成熟度.
[Abstract]:Aim: to observe the effects of granulocyte-macrophage colony stimulating factor (granulocyte-macrophage colonystimulating factor,GM-CSF) combined with interleukin-4 (interleukin,IL-4) on the morphology, purity and cell number of murine bone marrow-derived dendritic cells (dendritic cell,DC), and to explore the optimal conditions for the induction and culture of dendritic cells. Methods: the mouse bone marrow mononuclear cells were isolated and purified, and the morphologic and phenotypic observation and activation of allogeneic lymphocytes were performed by adding different concentration gradient GM-CSF and quantitative IL-4, respectively. Results: the number, purity and maturity of bone marrow-derived dendritic cells increased with the increase of GM-CSF concentration in a certain concentration range. Too high concentration of GM-CSF resulted in inhibitory effect. Therefore, it can be inferred that the ideal range of GM-CSF concentration should be 10-20 ng/mL.. Conclusion: the suitable concentration of GM-CSF combined with IL-4 can enhance the ability of mouse bone marrow mononuclear cells to differentiate into dendritic cells. The rational use of two kinds of cytokines can increase the number, purity and maturity of dendritic cells.
