【摘要】：駱駝科動物的血清中存在一種只含重鏈的同型二聚體(homodimer)構(gòu)成的特殊抗體,這類抗體由1個重鏈可變區(qū)和2個恒定區(qū)(CH2和CH3)組成,缺少CH1和輕鏈。由該類抗體重鏈可變區(qū)篩選制備而得到的抗體稱為單域抗體。單域抗體較常規(guī)抗體具有相對分子質(zhì)量小、熱穩(wěn)定性強(qiáng)、易通過血腦屏障等優(yōu)點(diǎn),越來越多地應(yīng)用于生物探測、診斷、制藥等領(lǐng)域。本文主要闡述了單域抗體的結(jié)構(gòu)特征、理化性質(zhì)及其在生物醫(yī)學(xué)領(lǐng)域的應(yīng)用。
[Abstract]:In the serum of Camelids, there is a special antibody composed of a homotypic dimer (homodimer) containing only heavy chain, which consists of a variable region of heavy chain and two constant regions (CH2 and CH3), and lacks CH1 and light chain. The antibody prepared by screening the heavy chain variable region of this kind of antibody is called single domain antibody. Compared with conventional antibodies, single-domain antibodies have the advantages of low molecular weight, strong thermal stability and easy to pass through the blood-brain barrier, so they are more and more used in biological detection, diagnosis, pharmaceutical and other fields. In this paper, the structure, physical and chemical properties of single-domain antibody and its application in biomedical field are reviewed.
