【摘要】：目的：觀察羊膜負(fù)載脂肪來(lái)源干細(xì)胞修復(fù)大鼠全層皮膚缺損。 方法：36只SD-大鼠隨機(jī)分成3組,每組12只,每只大鼠背部脊柱兩側(cè)分別設(shè)計(jì)1個(gè)全層皮膚缺損圓形創(chuàng)面(直徑2cm)。A組：將用CM-DiⅠ標(biāo)記的第3代脂肪來(lái)源干細(xì)胞接種到人脫細(xì)胞基質(zhì)羊膜共培養(yǎng)3天后覆蓋于創(chuàng)面；B組：以單純無(wú)種植細(xì)胞的人脫細(xì)胞基質(zhì)羊膜覆蓋于創(chuàng)面；C組：不予以處理。B組、C組作為對(duì)照組。觀察移植后3d、7d、10d 14d創(chuàng)面大小變化,創(chuàng)面愈合時(shí)間,切取創(chuàng)面組織行HE染色觀察肉芽組織生長(zhǎng)及上皮再生等；切取各組術(shù)后7d大鼠創(chuàng)面組織行熒光顯微鏡觀察。 結(jié)果：A組第14d創(chuàng)面已完全上皮化,全部愈合,創(chuàng)面閉合指數(shù)(wound closure index,WCI)為(100±0)%；B組和C組創(chuàng)面均未完全上皮化,其創(chuàng)面閉合指數(shù)分別為(95.05±2.65)%和(86.64±3.86)%。比較各組第3d、7d、10d、14d的WCI,A組較B組、A組較C組及B組較C組差別均有統(tǒng)計(jì)學(xué)意義(P0.01)。術(shù)后第7d,A組創(chuàng)面組織切片在熒光顯微鏡下可追蹤到ADSCs. 結(jié)論：羊膜負(fù)載脂肪來(lái)源干細(xì)胞能促進(jìn)大鼠全層皮肽缺損創(chuàng)面愈合。
