低氧條件對(duì)小鼠ADSCs膜片形成及其生物學(xué)特性的影響

【摘要】:目的:觀察低氧條件對(duì)小鼠脂肪間充質(zhì)干細(xì)胞(ADSCs)膜片生物學(xué)性能的影響。方法:分離培養(yǎng)C57/BL小鼠ADSCs并用Vit C法構(gòu)建細(xì)胞膜片;氯化鈷(CoCl_2)模擬體內(nèi)低氧環(huán)境,q PCR和Western Blot檢測(cè)HIF-1α表達(dá);組織學(xué)染色、透射電鏡觀察膜片形成;q PCR檢測(cè)ADSCs干性及膜片分泌功能相關(guān)基因的表達(dá)。結(jié)果:成功構(gòu)建ADSCs膜片;HIF-1α的表達(dá)隨CoCl_2濃度的升高而升高;低氧條件下,形成細(xì)胞膜片較薄,但膜片內(nèi)細(xì)胞連接更緊密;低氧環(huán)境可增強(qiáng)ADSCs干性,促使ADSCs膜片合成更多黏附分子、分泌更多保護(hù)性生長(zhǎng)因子。結(jié)論:低氧條件更利于ADSCs膜片構(gòu)建、存活和生物學(xué)功能維持。
[Abstract]:AIM: To observe the effects of hypoxia on the biological properties of adipose-derived mesenchymal stem cells (ADSCs) membrane in mice. Methods: ADSCs from C57/BL mice were isolated and cultured, and the cell membrane was constructed by Vit C method. Results: ADSCs membrane was successfully constructed, the expression of HIF-1a increased with the increase of CoCl_2 concentration, the formation of cell membrane was thinner under hypoxic condition, but the membrane cells were more closely connected; hypoxic environment could enhance the dryness of ADSCs, and promote the synthesis of more adhesion molecules and secretion of ADSCs membrane. CONCLUSION: Hypoxia is more conducive to the construction, survival and biological function of ADSCs.
