雙歧桿菌、乳酸桿菌在SD大鼠腸道動力研究

【摘要】:目的 探討雙歧桿菌與乳酸桿菌在SD大鼠的腸道動力及肥胖對其影響. 方法 利用羧基熒光素二乙酸琥珀酰亞胺酯(CFDA-SE)分子探針標(biāo)記雙歧桿菌、乳酸桿菌,以每只4ml×109CFU/ml灌喂肥胖及普飼組SD大鼠,分別于灌喂后0,2,4,8,12,24,48,72,96小時取大便,灌喂2,4,8,12,24小時取近端小腸、中段小腸、遠(yuǎn)端小腸、結(jié)腸等腸段,在熒光顯微鏡下觀察標(biāo)記菌數(shù)量。 結(jié)果 1.肥胖組雙歧桿菌2、4、24 h,乳酸桿菌2、12、24 h,大便細(xì)菌排出量較普飼組低(P0.05); 2.雙歧桿菌在肥胖組4h,普飼組2、4、8、24 h,大便細(xì)菌排出量與乳酸桿菌不同(P0.05); 3.肥胖組雙歧桿菌近端小腸在2、12 h,中段小腸在2、8 h,遠(yuǎn)端小腸在2、4、8、12h,結(jié)腸在2、4、12、24 h細(xì)菌在腸道的分布與普飼組不同(P0.05); 4.肥胖組乳酸桿菌中段小腸在2h,遠(yuǎn)端小腸在8、12、24 h,結(jié)腸在2、12、24 h與普飼組不同(P0.05). 結(jié)論 1.雙歧桿菌、乳酸桿菌在大鼠腸道動力不同:雙歧桿菌較乳酸桿菌腸道運動更快,大便細(xì)菌排出迅速;雙岐桿菌主要密集在結(jié)腸,而乳酸桿菌則能長時間停留在遠(yuǎn)端小腸及結(jié)腸; 2.肥胖對雙歧桿菌、乳酸桿菌的腸道動力有一定的影響:肥胖延緩細(xì)菌的腸道運動,使細(xì)菌在小腸內(nèi)長時間停留,大便細(xì)菌排出延遲,高峰推后,細(xì)菌排出波改變,排出量減少。
[Abstract]:Objective to investigate the intestinal motility of Bifidobacterium and Lactobacillus in SD rats and the effect of obesity. Methods Bifidobacterium and Lactobacillus were labeled with carboxyl fluorescein disuccinimide (CFDA-SE) molecular probe. Obese SD rats and SD rats were fed with 4ml 脳 109CFU/ml for 7296 hours. The proximal small intestine, middle small intestine, distal small intestine, colon and other intestinal segments were taken for 24 hours by perfusion. The number of labeled bacteria was observed under fluorescence microscope. Result 1. The excretion of fecal bacteria in obesity group was lower than that in general feeding group (P0.05). Bifidobacterium excretion was different from that of Lactobacillus in obesity group for 4 h and in general feeding group for 824 h (P0.05). The distribution of bacteria in the proximal small intestine of obesity group was different from that in the general feeding group (P0.05), the distribution of bacteria in the proximal small intestine was 2h, the middle intestine was 2h, the distal intestine was 812h, and the colon was 24 h. The middle small intestine of the obese group was at 2 hours, the distal small intestine was at 812 hours, and the colon was at 21: 12 hours, which was different from that of the general feeding group (P0.05). Conclusion 1. Bifidobacterium and Lactobacillus have different intestinal motility in rats: bifidobacterium moves faster than Lactobacillus and defecate bacteria excrete rapidly, bifidobacterium is mainly concentrated in colon, while Lactobacillus can stay in distal intestine and colon for a long time. 2. Obesity has a certain effect on the intestinal motility of bifidobacterium and lactobacillus: obesity delays the intestinal movement of bacteria, makes bacteria stay in the small intestine for a long time, delays fecal bacteria excretion, delays the peak, changes the bacterial excretion wave and reduces the excretion quantity.
