雌激素對(duì)絕經(jīng)后不同鼠齡大鼠血清一氧化氮水平的影響
[Abstract]:Objective to study the effect of estrogen on serum nitric oxide (NO) content in postmenopausal rats by establishing menopausal models of different ages. Effects of aortic endothelial nitric oxide synthase (eNOS) mRNA) expression and changes of aortic histomorphology. Methods Thirty young (3-month-old) female wistar rats and 30 aged (18-20 months) female wistar rats were randomly divided into three groups. There were 10 rats in each group: sham operation group (Sham group), ovariectomized group (OVX group), ovariectomized estradiol benzoate supplement group (OVE group) .Sham group: sham group was given 0.04m1 saline every 3 days; In OVE group, 10ug estradiol benzoate was injected intramuscularly every 3 days after removal of bilateral ovaries. After 4 months, the rats were killed and serum no levels were measured. The expression of eNOS mRNA in aorta and the histomorphologic changes of aorta were observed. Result 1. The serum no level of OVX group in young Sham group was significantly higher than that of OVX group in elderly Sham group. After ovariectomy, the serum no level in OVX group was significantly lower than that in Sham group (p 0.05), but there was no significant difference in aged OVX group. After estrogen supplementation, the serum no level in the young and elderly OVE group was significantly higher than that in the OVX group. The expression of aortic eNOS mRNA in the young Sham group was higher than that in the elderly Sham group. However, there was no significant difference in the expression of aortic eNOS mRNA between the young OVX group and the old OVX group. After ovariectomized, the expression of aortic eNOS mRNA in the young OVX group was significantly lower than that in the Sham group (p 0.05), and after estrogen supplementation, the expression of eNOS mRNA in the aorta of the young OVE group was significantly higher than that in the OVX group. There was no significant difference in the expression of eNOS mRNA in aorta between OVX group and OVX group in aged rats. Conclusion 1. In young postmenopausal rats, estrogen treatment may increase the level of serum nitric oxide (NO) by up-regulating the expression of endothelial nitric oxide synthase (eNOS) mRNA). In elderly postmenopausal rats, the increase in serum nitric oxide (NO) synthesis after estrogen therapy is not achieved by up-regulating the expression of endothelial nitric oxide synthase (eNOS) mRNA). The level of serum nitric oxide (NO) and the expression of endothelial nitric oxide synthase (eNOS) mRNA) decreased with age.
