α-2b干擾素對(duì)乙型肝炎模型鴨肝組織炎癥和肝纖維化的治療作用

【摘要】:目的:通過(guò)鴨乙型肝炎病毒(DHBV)陽(yáng)性血清注射建立鴨乙型肝炎模型,探討α-2b干擾素治療DHBV所致鴨肝炎癥及肝纖維化的作用,為干擾素的臨床應(yīng)用提供依據(jù)。方法:10日齡北京麻鴨40只隨機(jī)分為對(duì)照組(6只)和感染組(34只),感染組使用DHBV陽(yáng)性血清1周內(nèi)3次注射的方法,制備鴨乙型肝炎模型。2周后,采用斑點(diǎn)雜交法檢測(cè)DHBV-DNA水平確認(rèn)感染鴨共32只,隨機(jī)將感染動(dòng)物分為模型對(duì)照組(10只)、低劑量及高劑量α-2b干擾素組(各11只),α-2b干擾素組鴨分別肌肉注射低劑量(1×105 U·100g-1)和高劑量(5×105 U·100g-1)α-2b干擾素治療。30d治療結(jié)束后,采用斑點(diǎn)雜交法檢測(cè)各組鴨血清DHBV-DNA水平,檢測(cè)天門(mén)冬氨酸氨基轉(zhuǎn)移酶(AST)、谷氨酸氨基轉(zhuǎn)移酶(ALT)和肝纖維化指標(biāo)層黏蛋白(LN)及Ⅲ型前膠原肽(PCⅢ)水平,取肝組織檢測(cè)白細(xì)胞介素6(IL-6)、白細(xì)胞介素12(IL~(-1)2)和腫瘤壞死因子α(TNF-α)水平,觀(guān)察組織病理形態(tài)表現(xiàn)。結(jié)果:與造模前比較,造模后鴨血清DHBV-DNA水平明顯升高,表明造模成功。α-2b干擾素治療結(jié)束后,與模型組比較,低和高劑量α-2b干擾素組鴨血清DHBV-DNA水平明顯下降(P0.05),血清中AST和ALT水平降低(P0.05),其中高劑量α-2b干擾素組LN及PCⅢ型水平明顯降低(P0.05),肝組織中IL-6和TNF-α水平明顯降低(P0.05),IL~(-1)2水平無(wú)明顯變化(P0.05)。組織病理形態(tài)表現(xiàn),與模型組比較,低和高劑量α-2b干擾素組鴨肝組織中纖維化明顯減輕。結(jié)論:α-2b干擾素可以減輕DHBV所致的鴨肝組織炎癥和肝纖維化,本研究為干擾素的進(jìn)一步開(kāi)發(fā)和臨床應(yīng)用提供了理論依據(jù)。
[Abstract]:Objective: to establish duck hepatitis B model by injecting duck hepatitis B virus (DHBV) positive serum to investigate the effect of 偽 -2b interferon on duck liver inflammation and hepatic fibrosis induced by DHBV, and to provide evidence for clinical application of interferon. Methods 40 10 day old Beijing duck were randomly divided into control group (6) and infected group (34). The infected group was injected with DHBV positive serum 3 times within one week to prepare duck hepatitis B model after .2 weeks. The DHBV-DNA level of 32 infected ducks was detected by dot blot hybridization. The infected animals were randomly divided into model control group (n = 10), low dose interferon group (n = 11) and high-dose interferon 偽 -2b group (n = 11 each). The ducks of 偽 -2b interferon group were injected intramuscularly with low dose (1 脳 10 5 U 100g-1) and high dose (5 脳 10 5 U 100g-1) 偽 -2b interferon for 30 days.
