新疆中哈邊境地區(qū)分離到遠(yuǎn)東型和西伯利亞型蜱傳腦炎病毒

【摘要】:目的研究新疆中哈邊境阿拉套山夏爾西里自然保護(hù)區(qū)蜱傳腦炎疫源地病原的基因型及生物學(xué)特征。方法采用布旗法采集蜱,活蜱保存或液氮凍存;采用BALB/c小鼠與BHK-21細(xì)胞進(jìn)行蜱傳腦炎病毒分離培養(yǎng);采用RT-PCR擴(kuò)增蜱傳腦炎病毒遠(yuǎn)東型FE和西伯利亞型S特異基因片段并測定其序列。結(jié)果從新疆中哈邊境阿拉套山夏爾西里自然保護(hù)區(qū)全溝硬蜱和森林革蜱中分離出16株蜱傳腦炎病毒株,通過對擴(kuò)增基因序列比對分析,明確其中13株為遠(yuǎn)東型,3株為西伯利亞型。結(jié)論從新疆中哈邊境阿拉套山夏爾西里自然保護(hù)區(qū)分離到遠(yuǎn)東型和西伯利亞型蜱傳腦炎病毒,該地區(qū)為兩種亞型病毒共存的蜱傳腦炎自然疫源地。
[Abstract]:Objective to study the genotypes and biological characteristics of tick-borne encephalitis foci in Alatao Mountain, Xinjiang. Methods ticks were collected by cloth flag method, stored by living ticks or frozen by liquid nitrogen, and isolated and cultured by BALB/c mice and BHK-21 cells. RT-PCR was used to amplify and sequence the FFE and S specific genes of tick-borne encephalitis virus. Results Sixteen tick-borne encephalitis virus strains were isolated from Chalxi Nature Reserve in Alatao Mountain, Xinjiang, China. The results showed that 16 tick-borne encephalitis virus strains were analyzed by sequence alignment of amplified genes. 13 of them were of far East type and 3 of them were Siberia type. Conclusion far East and Siberian tick-borne encephalitis viruses were isolated from Alatao Mountain Sherri Nature Reserve in the Sino-Kazak border of Xinjiang. This area is the natural focus of tick-borne encephalitis in which two subtypes of viruses coexist.
