基于斑馬魚急性模型的抗缺氧活性成分篩選評(píng)價(jià)

【摘要】:目的建立斑馬魚的急性缺氧模型,并對(duì)五環(huán)三萜類化合物的抗缺氧活性進(jìn)行初篩。方法 64只斑馬魚放置低氧操作箱中建立斑馬魚的急性缺氧模型,并隨機(jī)分成空白組、陽(yáng)性對(duì)照組[槲皮素(QT)]、積雪草酸(AA)組、羥基積雪草酸(MA)組、積雪草苷(AS)組、羥基積雪草苷(MS)組、熊果酸(UA)組、甘草次酸(GA)組,每組8只。實(shí)驗(yàn)組和陽(yáng)性對(duì)照組所加藥物的終濃度均為2.5μg·mL~(-1),空白組為加入相同劑量的20%PEG-400溶液。比較各組斑馬魚的生理狀態(tài)和半數(shù)死亡(4只)所用時(shí)間。結(jié)果斑馬魚的缺氧條件為20℃,3%的氧氣濃度。空白組、陽(yáng)性對(duì)照組、AA組、MA組、AS組、MS組、UA組、GA組的半數(shù)死亡所用時(shí)間分別為(126.22±4.92),(166.98±3.30),(157.02±4.62),(181.02±3.54),(118.98±3.78),(121.98±1.98),(127.98±2.82),(129.48±5.94)min,除AS組和GA組外,其他組斑馬魚半數(shù)死亡時(shí)間與空白組比較,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論用斑馬魚急性缺氧模型對(duì)潛在抗缺氧活性物質(zhì)的篩選評(píng)價(jià)建模容易、結(jié)果可靠;三萜類苷元(AA、MA、UA和GA)具有抗缺氧作用,且烏蘇烷型三萜(AA、MA和UA)的抗缺氧作用更明顯;三萜類皂苷(AS、MS)不具有抗缺氧活性。

Methods the acute hypoxia model of zebrafish was established by placing 64 zebrafish in hypoxic box. The model was randomly divided into three groups: positive control group [quercetin (QT)], (AA) group, (MA) group and (AS) group. Hydroxy asiaticoside (MS) group, ursolic acid (UA) group, glycyrrhetinic acid (GA) group, 8 rats in each group. The final concentration of the drug was 2.5 渭 g mL ~ (-1) in the experimental group and the positive control group, and the same dose of 20%PEG-400 solution was added in the blank group.
