高果糖飲食誘導(dǎo)大鼠代謝綜合征的模型研究

【摘要】:目的:建立高果糖飲食誘導(dǎo)大鼠代謝綜合征模型。方法:將周齡為8周的雄性SD大鼠隨機分成2組,對照組(n=15)喂飲普通凈水,果糖組(n=33)喂飲15%果糖水,共8周。于喂飲果糖前后測定各組大鼠的體重、24小時飲食、12小時尿量、血壓、空腹血糖和血液生化指標。喂飲果糖8周后,通過胰島素耐量試驗和高胰島素-正葡萄糖鉗夾技術(shù)比較各組大鼠胰島素敏感性。結(jié)果:喂飲果糖導(dǎo)致動物出現(xiàn)了多飲多尿少食的現(xiàn)象。與對照組大鼠相比,果糖組大鼠在喂飲果糖6-8周后,產(chǎn)生了明顯的高血壓、高脂血癥、胰島素抵抗及高胰島素血癥等癥狀。8周的果糖喂飲并未導(dǎo)致果糖組大鼠出現(xiàn)明顯的肥胖和高血糖,但ITT和糖鉗實驗證明,果糖組大鼠出現(xiàn)了明顯的胰島素抵抗。結(jié)論:成功建立高果糖飲食誘導(dǎo)大鼠代謝綜合征模型,為進一步研究代謝綜合征的病理生理機制和開發(fā)預(yù)防和治療代謝綜合征的藥物提供基礎(chǔ)。
[Abstract]:Objective: to establish a rat model of metabolic syndrome induced by high fructose diet. Methods: male Sprague-Dawley rats aged 8 weeks were randomly divided into two groups: the control group (n = 15) was fed with normal purified water, and the fructose group (n ~ (33) was fed with 15% fructose water for 8 weeks. Before and after drinking fructose, the body weight of rats in each group was measured, and the urine volume, blood pressure, fasting blood glucose and blood biochemical indexes were measured. After fed fructose for 8 weeks, insulin sensitivity was compared by insulin tolerance test and hyperinsulin-glucose clamp technique. Results: feeding fructose led animals to drink more urine and less food. Compared with the control group, the fructose group produced significant hypertension and hyperlipidemia after 6-8 weeks of drinking fructose. 8 weeks of fructose feeding with insulin resistance and hyperinsulinemia did not result in obvious obesity and hyperglycemia in fructose group, but ITT and glucose clamp experiments showed that fructose group had obvious insulin resistance. Conclusion: the rat model of metabolic syndrome induced by high fructose diet is successfully established, which provides a basis for further study on the pathophysiological mechanism of metabolic syndrome and the development of drugs for prevention and treatment of metabolic syndrome.
