洋苷菊正丁醇提取物對(duì)哮喘模型小鼠的作用機(jī)理

【摘要】:目的研究維藥洋苷菊提取物對(duì)支氣管哮喘小鼠模型的治療作用,探討其抗哮喘的可能機(jī)制。方法 KM種小鼠60只隨機(jī)分為6組:正常對(duì)照組,模型組,陽性對(duì)照組,洋苷菊正丁醇提取物低、中、高劑量組。采用卵蛋白(OVA)腹腔注射致敏與霧化吸入激發(fā)制作哮喘模型;致敏第15天開始每次霧化激發(fā)前1 h給予洋苷菊治療,連續(xù)4周。末次給藥后1 h,檢測(cè)各組小鼠的肺功能及增強(qiáng)的呼吸間歇;取肺泡灌洗液(BALF)測(cè)定細(xì)胞計(jì)數(shù),頸動(dòng)脈取血,離心取血清測(cè)定白介素IL-2、IL-10及IL-12含有量;肺組織勻漿離心取上清液檢測(cè)超氧化物歧化酶(SOD)和丙二醛(MDA)水平;HE染色觀察肺組織病理改變。結(jié)果洋甘菊正丁醇提取物可改善哮喘小鼠肺功能;與正常對(duì)照組相比,模型組增強(qiáng)的呼吸間歇(Penh)明顯升高(P0.01);與模型組相比,各給藥組Penh顯著降低小鼠(P0.05或P0.01),BALF嗜酸性粒細(xì)胞計(jì)數(shù)(EOS)及肺組織MDA各組水平均有一定程度的降低(P0.05或P0.01);各治療組肺組織SOD及血清中IL-2、IL-10、IL-12含有量明顯升高(P0.05,P0.01)。結(jié)論洋甘菊正丁醇提取物具有明顯的平喘作用;洋甘菊正丁醇提取物對(duì)肺泡灌洗液中EOS、肺組織MDA水平有一定降低作用;對(duì)血清IL-2、IL-10、IL-12及肺組織SOD水平有一定升高作用;洋甘菊正丁醇提取物通過改善哮喘小鼠肺功能,降低Penh,改善哮喘小鼠氣道高反應(yīng)性。
[Abstract]:Objective to study the therapeutic effect of the extract of guerarin on asthma model in mice, and to explore the possible mechanism of its anti-asthma. Methods 60 km mice were randomly divided into 6 groups: normal control group, model group, positive control group, low, middle and high dose group. The asthmatic model was induced by intraperitoneal injection of ovalbumin (OVA) and inhalation of atomization, and the rats were treated with guanosin 1 hour before each atomization on the 15th day of sensitization for 4 weeks. At 1 hour after the last administration, lung function and enhanced respiratory interval were detected in each group, cell count was determined by (BALF) in alveolar lavage fluid, blood was taken from carotid artery, IL-10 and IL-12 content in serum were measured by centrifugation. Superoxide dismutase (SOD) and malondialdehyde (MDA) (MDA) were detected by HE staining in supernatant of lung tissue homogenate. Results the pulmonary function of asthmatic mice was improved by n-butanol extract from chamomile, the (Penh) of the model group was significantly higher than that of the normal control group (P0.01), and that of the model group was higher than that of the model group. The eosinophilic granulocyte count (EOS) and the level of MDA in lung tissue were significantly decreased in each treatment group (P0.05 or P0.01), and the contents of IL-2IL-10 IL-12 in lung tissue and serum of each treatment group were significantly increased (P0.05, P0.01). Conclusion the n-butanol extract of Chamomile has obvious antiasthmatic effect, the extract of n-butanol can decrease the level of MDA in alveolar lavage fluid, increase the level of IL-10 IL-12 in serum and SOD in lung tissue. Chamomile n-butanol extract improved airway hyperresponsiveness of asthmatic mice by improving lung function, reducing Penhs and improving airway hyperresponsiveness of asthmatic mice.
