腹腔感染大鼠腸粘膜通透性改變與E-cadherin蛋白表達(dá)的關(guān)系
[Abstract]:Objective to observe the changes of intestinal permeability after intestinal barrier injury in rats with abdominal infection and its correlation with injury degree and expression of E-cadherin protein. Methods Forty healthy adult male Wistar rats were randomly divided into two groups: control group (simple caesarean section only) and celiac infection (abdominal infection model made by cecal ligation and perforation) 12 h after operation, 8 rats in each group were killed. The ileal 2cm from ileocecal 10cm was examined by pathological examination (including routine examination, intestinal epithelium injury index) and plasma D-lactic acid content at the corresponding time point. The E-cadherin protein level in intestinal mucosa was measured by Western blot method. Results (1) after 12 hours of intraperitoneal infection, obvious changes of intestinal barrier began to appear: intestinal mucosal injury score increased, plasma D- lactic acid content increased significantly (P0.01) and 24 h injury reached its peak: intestinal mucosal injury score was the highest. Plasma D- lactic acid content was significantly increased (P0.05), then intestinal barrier gradually recovered, but the injury degree of 48h group was still significantly higher than that of the control group (P0.01). There was a significant correlation between the plasma D- lactic acid content and the injury index (r = 0.489g, P0.01), compared with the control group. The expression of E-cadherin protein in the experimental group was significantly lower than that in the control group (P0.05). The level of E-cadherin protein in the 48h group was significantly higher than that in the 24h group and 36h group (P < 0.05). There was no significant difference between the two groups (P0.05). There was a significant correlation between plasma D-lactic acid level and E-cadherin protein content (r-0. 605, P 0. 01). Conclusion under the condition of abdominal infection, the intestinal barrier was seriously damaged, and the change of intestinal mucosal permeability was significantly correlated with the degree of intestinal barrier injury and the level of E-cadherin protein.
