【摘要】：目的觀察腹腔感染大鼠腸屏障損傷后腸通透性的變化,及其與損傷程度和E-cadherin蛋白表達(dá)的相關(guān)性。 方法健康成年雄性Wistar大鼠40只,隨機(jī)分為空白對(duì)照組(僅行單純剖腹手術(shù))和腹腔感染(采用盲腸結(jié)扎穿孔法制作腹腔感染模型)術(shù)后12h、24、36、48h組,每組8只,處死后,距回盲部10cm處取回腸2cm進(jìn)行病理檢查(包括常規(guī)檢查、測(cè)定腸上皮損傷指數(shù))及測(cè)定相應(yīng)時(shí)間點(diǎn)血漿D-乳酸含量,應(yīng)用Western blot方法測(cè)定腸粘膜E-cadherin蛋白的水平。 結(jié)果①腹腔感染12h后腸屏障開(kāi)始出現(xiàn)明顯損傷性改變：腸粘膜損傷評(píng)分增加,血漿D-乳酸含量明顯增加(P0.01)；24h損傷達(dá)到高峰：腸粘膜損傷評(píng)分最大,血漿D-乳酸含量明顯增高(P0.05)；隨后腸屏障逐漸恢復(fù),但48h組損傷程度仍顯著高于對(duì)照組(P0.01),血漿D-乳酸含量與損傷指數(shù)具有顯著相關(guān)性(r=0.489,P0.01)。②與對(duì)照組比較,實(shí)驗(yàn)組E-cadherin蛋白表達(dá)量降低(P0.05)：12h組、48h組的E-cadherin蛋白水平分別與24h組和36h組比較均有統(tǒng)計(jì)學(xué)意義(P＜0.05)：24h組和36h組比較無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。血漿D-乳酸水平與E-cadherin蛋白含量具有明顯相關(guān)性(r=-0.605,P0.01)。 結(jié)論腹腔感染狀態(tài)下,腸屏障嚴(yán)重受損,腸粘膜通透性的改變與腸屏障損傷程度及E-cadherin蛋白水平均具有顯著的相關(guān)性。
[Abstract]:Objective to observe the changes of intestinal permeability after intestinal barrier injury in rats with abdominal infection and its correlation with injury degree and expression of E-cadherin protein. Methods Forty healthy adult male Wistar rats were randomly divided into two groups: control group (simple caesarean section only) and celiac infection (abdominal infection model made by cecal ligation and perforation) 12 h after operation, 8 rats in each group were killed. The ileal 2cm from ileocecal 10cm was examined by pathological examination (including routine examination, intestinal epithelium injury index) and plasma D-lactic acid content at the corresponding time point. The E-cadherin protein level in intestinal mucosa was measured by Western blot method. Results (1) after 12 hours of intraperitoneal infection, obvious changes of intestinal barrier began to appear: intestinal mucosal injury score increased, plasma D- lactic acid content increased significantly (P0.01) and 24 h injury reached its peak: intestinal mucosal injury score was the highest. Plasma D- lactic acid content was significantly increased (P0.05), then intestinal barrier gradually recovered, but the injury degree of 48h group was still significantly higher than that of the control group (P0.01). There was a significant correlation between the plasma D- lactic acid content and the injury index (r = 0.489g, P0.01), compared with the control group. The expression of E-cadherin protein in the experimental group was significantly lower than that in the control group (P0.05). The level of E-cadherin protein in the 48h group was significantly higher than that in the 24h group and 36h group (P < 0.05). There was no significant difference between the two groups (P0.05). There was a significant correlation between plasma D-lactic acid level and E-cadherin protein content (r-0. 605, P 0. 01). Conclusion under the condition of abdominal infection, the intestinal barrier was seriously damaged, and the change of intestinal mucosal permeability was significantly correlated with the degree of intestinal barrier injury and the level of E-cadherin protein.
