丘腦腹內(nèi)側(cè)核高頻電刺激改善帕金森病模型大鼠前肢使用不對稱行為的研究
[Abstract]:Objective to investigate whether high frequency stimulation of ventromedial nucleus of thalamus can improve the asymmetric behavior of forelimb in Parkinson's disease model rats. Methods 33 SD rats were randomly divided into experimental group (n = 22) and control group (n = 11). The stimulation electrode was implanted into the ventromedial nucleus of the right thalamus by stereotactic method and the 6-hydroxydopamine cannula was injected into the right medial forebrain tract. The right medial forebrain of the experimental group was injected with 12 渭 g of 6-OHDAand the control group was injected with the same volume of normal saline. The antebrachial asymmetry test was performed on rats before, after and after DBS administration. The damage of substantia nigra dopaminergic neurons was detected by tyrosine hydroxylase immunohistochemical staining. The tip position of the stimulation electrode was determined by Nissl staining. Results Fifteen successful PD rat models were obtained from 22 rats. The asymmetry index of model rats increased significantly (P0. 000). After high frequency VM-DBS, the asymmetry index decreased significantly (P 0. 009), but there was no significant difference (P 0. 214). There was no significant change in asymmetry index after low frequency VM-DBS, but there was no significant difference (P0. 036) between the two groups when VM-DBS was not given after the damage (P0. 4), but significant difference was found between the two groups (P0. 036). Conclusion High frequency electrical stimulation of ventromedial nucleus of thalamus can improve the asymmetric behavior of the forelimb of 6-hydroxydopamine hemiparkinsonism rats. The ventromedial nucleus of thalamus may be a potentially effective target nucleus for deep brain stimulation in Parkinson's disease.
