益腎通癃膠囊對前列腺增生模型小鼠生殖激素水平的影響

【摘要】:目的:探討益腎通癃膠囊對良性前列腺增生(BPH)模型小鼠生殖激素水平的影響。方法:對SPF雄性小鼠皮下注射丙酸睪酮5mg/(kg·d),連續(xù)3周,待造模成功后分為6組:正常對照組,模型組,癃閉舒膠囊組,益腎通癃膠囊高、中、低劑量組,每組10只。益腎通癃膠囊高、中、低劑量組分別用1.2,0.6,0.3 g/kg益腎通癃膠囊混懸液灌胃,癃閉舒膠囊組用0.45 g/kg癃閉舒膠囊混懸液灌胃,正常對照組及模型組給予同體積蒸餾水。給藥8周后處死小鼠,稱取前列腺濕重并計算前列腺指數(shù)(PI),測量小鼠血清中睪酮(T)、雌二醇(E2)的水平、E2/T比值,光鏡下觀察小鼠前列腺組織的形態(tài)變化。結果:與模型組比較,益腎通癃膠囊中、高劑量組及癃閉舒膠囊組均可降低模型小鼠前列腺濕重、PI指數(shù)(P均0.01)。各組小鼠血清T、E2、E2/T水平:正常對照組:(1.73±0.02)ng/ml、(73.08±1.03)pg/ml、42.30±0.53;模型組:(3.86±0.02)ng/ml,(145.79±0.88)pg/ml,37.76±0.25;癃閉舒膠囊組:(2.47±0.02)ng/ml,(95.87±0.47)pg/ml,38.80±0.13;益腎通癃膠囊高劑量組:(2.75±0.03)ng/ml,(107.11±0.61)pg/ml,38.92±0.36;益腎通癃膠囊中劑量組:(2.77±0.02)ng/ml,(112.16±0.82)pg/ml,40.56±0.29;益腎通癃膠囊低劑量組:(2.91±0.03)ng/ml,(112.68±0.77)pg/ml,38.80±0.42,各組與模型組相比,差異均具有統(tǒng)計學意義(P0.05或P0.01)。結論:益腎通癃膠囊可能通過降低BPH模型小鼠血清T、E2,升高E2/T比值,揭示了益腎通癃膠囊治療BPH的機制。
[Abstract]:Objective: To investigate the effect of Yishen Tong Long capsule (Yishen Tong Long capsule) on the level of reproductive hormone in mice with benign prostatic hyperplasia (BPH). Methods: SPF male mice were subcutaneously injected with testosterone propionate 5mg/ (kg. D) for 3 weeks. After the model was successful, the model group was divided into 6 groups: normal control group, model group, yishanshu capsule group, Yishen Tong Long capsule high, medium and low dose group, 10 rats in each group. Yishen Tong Long capsule high, medium and low dose group were fed with 1.2,0.6,0.3 g/kg Yishen Tong Long capsule suspension, respectively, and long Shu Capsule group was fed with 0.45 g/kg long long closed capsule suspension. The normal control group and the model group were given the same volume of distilled water. After 8 weeks of administration, the mice were killed and the prostatic index (PI) was measured and the mice were measured, and the mice were measured. The level of serum testosterone (T), estradiol (E2), E2/T ratio, and the morphological changes of the prostate tissue in mice were observed under light microscope. Results: compared with the model group, the high dose group and the long closed capsule group could reduce the wet weight of the prostate in the model mice and the number of PI (P 0.01). The serum T, E2, E2/T level of the mice in each group: (1.7 3 + 0.02) ng/ml, (73.08 + 1.03) pg/ml, 42.30 + 0.53, model group: (3.86 + 0.02) ng/ml, (145.79 + 0.88) pg/ml, 37.76 + 0.25, (2.47 + 0.02) ng/ml, (2.47 + 0.53) ng/ml, high dose group of Yishen Tong Long capsule: ng/ml /ml, (112.16 + 0.82) pg/ml, 40.56 + 0.29, Yishen Tong Long capsule low dose group: (2.91 + 0.03) ng/ml, (112.68 + 0.77) pg/ml, 38.80 + 0.42, the difference was statistically significant compared with the model group (P0.05 or P0.01). Conclusion: Yishen Tong Long capsule may reduce the serum T, E2, and E2/T ratio of BPH model mice, and reveal Yishen Tong Long glue The mechanism of the cysts for the treatment of BPH.
