沙門菌屬特異性檢測靶點(diǎn)堿基差異位點(diǎn)的識別及其血清型決定力

【摘要】:以美國國家生物技術(shù)信息中心(NCBI)已公布的28株沙門菌(14個(gè)血清型)全基因組序列為研究對象,對前期研究獲得的7個(gè)沙門菌屬特異性檢測靶點(diǎn)在不同血清型間的單核苷酸多態(tài)性進(jìn)行比較分析,結(jié)果表明,S9和S69為堿基差異位點(diǎn)最多的兩個(gè)靶點(diǎn),具有沙門菌分子血清分型的潛在能力。隨后,通過分析S9和S69在沙門菌不同血清型菌株中的差異位點(diǎn),分別繪制兩個(gè)血清分型靶點(diǎn)的差異位點(diǎn)表,從而獲得了這兩個(gè)靶點(diǎn)同時(shí)用于14個(gè)沙門菌血清型的快速分子血清分型的差異位點(diǎn)組合。在這些組合中,同一血清型具有相同的差異位點(diǎn),不同血清型可以通過差異位點(diǎn)得以區(qū)分。最后,采集食品樣品192份,用國標(biāo)方法獲得疑似沙門菌21株;利用血清分子分型靶點(diǎn)S9和S69進(jìn)行快速分型,并同時(shí)用傳統(tǒng)玻片凝集反應(yīng)鑒定方法進(jìn)行驗(yàn)證,兩種方法鑒定結(jié)果的符合率為100%。鑒定結(jié)果為:21株沙門菌共包括4個(gè)不同血清型,其中腸炎沙門菌10株、鼠傷寒沙門菌7株、雞沙門菌2株、紐波特沙門菌2株。基因組序列分析結(jié)果和分離株分型結(jié)果均表明,沙門菌特異分子檢測靶點(diǎn)S9和S69相結(jié)合具備沙門菌的分子血清分型能力,以差異位點(diǎn)表為基礎(chǔ)建立的血清分型方法有望替代傳統(tǒng)的玻片凝集血清分型這一繁雜步驟,從而降低沙門菌血清鑒定的時(shí)間與成本,為聚合酶鏈?zhǔn)椒磻?yīng)技術(shù)應(yīng)用于沙門菌分子血清分型提供新思路。
[Abstract]:The complete genome sequences of 28 Salmonella strains (14 serotypes) from the National Biotechnology Information Center (NCBI) were studied. The single nucleotide polymorphisms of 7 Salmonella specific detection targets among different serotypes were compared and analyzed. The results showed that S9 and S69 were the two targets with the most different base sites. It has the potential of Salmonella serotyping. Then, by analyzing the difference sites of S9 and S69 in different serotypes of Salmonella, the difference sites of the two serotyping targets were drawn. The difference site combinations of the two targets for rapid molecular serotyping of 14 Salmonella serotypes were obtained. In these combinations, the same serotype has the same differential loci, and different serotypes can be distinguished by differential loci. Finally, 192 food samples were collected and 21 strains of Salmonella suspected were obtained by national standard method, and rapid typing was carried out by using serum molecular typing sites S9 and S69, and verified by traditional slide agglutination reaction. The coincidence rate of the two methods was 100. The results showed that there were 4 different serotypes in 21 strains of Salmonella, including 10 strains of Salmonella enteritis, 7 strains of Salmonella typhimurium, 2 strains of Salmonella chickenii and 2 strains of Salmonella Newport. The results of genomic sequence analysis and isolate typing showed that S9 and S69, the specific molecular detection sites of Salmonella, had the ability of molecular serotyping of Salmonella. The serotyping method based on the differential locus table is expected to replace the traditional slide agglutination serotyping, thus reducing the time and cost of the identification of Salmonella serums. It provides a new idea for the application of polymerase chain reaction in Salmonella serotyping.
