下丘腦—海馬Ghrelin神經(jīng)元投射及海馬微量注射Ghrelin對大鼠攝食、體重及胃運動的影響

【摘要】:目的:觀察下丘腦室旁核(PVN)-海馬Ghrelin神經(jīng)元的投射;探討海馬微量注射Ghrelin對大鼠攝食、體重、胃運動的影響。 方法:采用熒光金(Fluorogold, FG)逆行追蹤結(jié)合熒光免疫組化方法,探討下丘腦PVN Ghrelin神經(jīng)元向海馬的投射,以及海馬神經(jīng)元向下丘腦PVN的投射;采用海馬核團微量注射Ghrelin,觀察清醒大鼠攝食、體重及胃運動的改變。 結(jié)果:海馬微量注射FG,7天后可見下丘腦PVN有大量標記FG的神經(jīng)元,且呈同側(cè)優(yōu)勢表達,結(jié)合Ghrelin熒光免疫組化實驗,發(fā)現(xiàn)部分下丘腦PVN FG神經(jīng)元有Ghrelin免疫陽性物的表達;下丘腦PVN微量注射FG,7天后可見海馬CA1、CA2-3區(qū)有大量FG標記的神經(jīng)元,且呈同側(cè)優(yōu)勢表達。海馬CA1區(qū)微量注射Ghrelin后,大鼠攝食和體重明顯增加(P0.05),胃運動明顯增強(P0.05),且呈顯著劑量依賴關(guān)系。 結(jié)論:下丘腦PVN-海馬有往返纖維投射,且呈同側(cè)優(yōu)勢投射;下丘腦PVN Ghrelin免疫陽性神經(jīng)元投向海馬CA1區(qū);海馬CA1區(qū)Ghrelin免疫陽性神經(jīng)元投向下丘腦PVN;海馬CA1區(qū)Ghrelin通過其受體參與大鼠攝食、體重和胃運動的調(diào)控。
[Abstract]:Aim: to observe the projections of hypothalamic paraventricular nucleus (PVN)-hippocampal ghrelin neurons and to investigate the effects of hippocampal microinjection of ghrelin on feeding weight and gastric motility in rats. Methods: the projections of hypothalamic PVN ghrelin neurons to hippocampus and hippocampal neurons to hypothalamic PVN were studied by using Fluorogold (FG) retrograde tracing and fluorescence immunohistochemistry. Hippocampal nuclei were microinjected with ghrelin to observe the changes of food intake, body weight and gastric motility in conscious rats. Results: a large number of neurons labeled with FG were found in hypothalamus PVN 7 days after microinjection of FGG, and the expression of FG was ipsilateral dominant. Combined with ghrelin fluorescence immunohistochemical experiment, ghrelin immunoreactivity was found in some hypothalamic PVN FG neurons. 7 days after microinjection of hypothalamic PVN, a large number of FG labeled neurons were found in hippocampal CA1P CA2-3 area, and showed ipsilateral dominant expression. After microinjection of ghrelin in hippocampal CA1 area, the feeding and body weight of the rats increased significantly (P0.05), and the gastric motility increased significantly (P0.05) in a dose-dependent manner. Conclusion: hypothalamic PVN- hippocampal neurons have homolateral projections, hypothalamic PVN ghrelin immunoreactive neurons turn to hippocampal CA1 region, hippocampal CA1 region ghrelin immunoreactive neurons into hypothalamic PVNs. Ghrelin in hippocampal CA1 region participates in the regulation of feeding, body weight and gastric motility through its receptor.
