東莨菪堿與苯扎氯銨干眼癥大鼠造模方法研究

【摘要】:為比較氫溴酸東莨菪堿皮下注射與苯扎氯銨滴眼制作干眼癥動物模型的各自特點,將健康SD大鼠隨機分為正常組、東莨菪堿加風組、苯扎氯銨組和吹風對照組。正常組不接受任何處理;東莨菪堿組皮下注射氫溴酸東莨菪堿注射劑;苯扎氯銨組以2mg/mL苯扎氯銨滴眼液每日3次滴眼;吹風組僅置于吹風裝置中每日12h。于試驗第3、7、10天檢查Schirmer I試驗(SIT)、淚膜破裂時間(BUT)、角膜熒光素染色(FLS)。結(jié)果表明,東莨菪堿組在第3、7、10天,SIT與正常組相比具有顯著差異(P0.05),第7、10天BUT和FLS與正常組相比具有顯著差異(P0.05);苯扎氯銨組在試驗第7、10天,SIT試驗和BUT與正常組相比有顯著差異(P0.05),BUT第10天差異達到最大,在第3、7、10天,FLS與正常組相比具有顯著差異(P0.05);組織病理學檢查東莨菪堿組淚腺較苯扎氯銨組明顯有腺上皮細胞萎縮;苯扎氯銨組大鼠角結(jié)膜上皮細胞明顯變薄,表層細胞排列紊亂,細胞數(shù)量顯著減少,并存在炎性細胞浸潤。提示造模10d氫溴酸東莨菪堿注射劑皮下注射配合吹風制作的干眼癥動物模型對大鼠淚腺及眼表上皮細胞影響顯著,伴有較輕程度的眼表損傷,更適合早期輕中度干眼造模;苯扎氯銨滴眼造模方法易造成眼表上皮損傷,適合短期重度干眼的造模。
[Abstract]:In order to compare the characteristics of scopolamine hydrobromide subcutaneous injection and benzalkonium chloride dropping eye to make dry eye animal model, healthy SD rats were randomly divided into normal group, scopolamine plus wind group, benzalkonium chloride group and blow control group. Scopolamine hydrobromide injection was injected subcutaneously in scopolamine group, 2 mg / mL benzalkonium chloride eye drops were used three times a day in the benzalkonium chloride group, and only 12 hours a day in the blowing group. Schirmer I test (sit), tear film rupture time (but) and corneal fluorescein staining (FLS) were examined on day 3. The results show that There was significant difference between scopolamine group and normal group on day 3 (P0.05), but and FLS on day 7 (P0.05), and that of sit and but on day 710 (P0.05) in benzalkonium chloride group (P0.05), and that of but on day 710 in anisolamine group was significantly higher than that in normal group (P0.05), and that of but in group B was significantly higher than that in group B on day 710 (P0.05), and that of but in group B was significantly higher than that in group B on day 710 (P0.05). The difference reached the maximum on the 10th day, There was significant difference between FLS and normal group on the 3rd 7th day (P0.05). Histopathological examination showed that the lacrimal gland of scopolamine group had obvious atrophy of glandular epithelium than that of benzalkonium chloride group, the epithelial cells of cornea conjunctiva of rats in benzalkonium chloride group were thinning obviously, and the surface cells were disordered. The number of cells decreased significantly and inflammatory cell infiltration occurred. It is suggested that the animal model of dry eye induced by injection of scopolamine hydrobromide injection and blowing air for 10 days has a significant effect on lacrimal gland and ocular surface epithelial cells, which is more suitable for early dry eye model. The method of eye modeling with benzalkonium chloride dripping is easy to cause ocular surface epithelium injury and is suitable for short term dry eye modeling.
