【摘要】：目的研究裕固族男性飲酒行為及其與相關(guān)遺傳學(xué)特征的關(guān)系,為飲酒相關(guān)疾病的防治提供科學(xué)依據(jù)。 方法(1)采用自評式問卷對肅南縣505名裕固族男性飲酒狀況進(jìn)行調(diào)查,通過單因素和多因素回歸分析探討飲酒行為的影響因素；(2)采用酒精依賴性診斷量表(ADS)篩選出酒精依賴者,比較灑精依賴人群和非酒精依賴人群血脂、肝功和腎功能以及乙醇脫氫酶3(ADH3)和乙醛脫氫酶2(ALDH2)基因多態(tài)型的分布�；蚨鄳B(tài)型的檢測采用聚合酶鏈?zhǔn)椒磻?yīng)-限制性片段長度多態(tài)性(PCR-RFLP)方法。 結(jié)果(1)裕固族男性飲酒率為56.83%,酒精依賴患病率為9.90%;Logistic回歸結(jié)果提示與飲酒行為有獨(dú)立相關(guān)關(guān)系的因素包括家庭收入、婚姻狀況、日吸煙量(正相關(guān)P0.05)、年齡、文化程度、職業(yè)、健康狀況(負(fù)相關(guān)P0.05)；與酒精依賴有獨(dú)立相關(guān)關(guān)系的因素包括年齡、家庭收入、婚姻狀況、日吸煙量、年飲酒量(正相關(guān)P0.05)、文化程度、職業(yè)、健康狀況(負(fù)相關(guān)P0.05);(2)飲酒量與血脂、肝腎功能指標(biāo)存在相關(guān)關(guān)系；(3)ALDH2*2等位基因的分布與飲酒量負(fù)相關(guān),裕固族男性ALDH2*1和ALDH2*2等位基因的分布頻率分別為：76.82%和33.18%:ADH3*1和ADH3*2等位基因的分布頻率分別為：87.87%和16.13%。 結(jié)論裕固族男性的飲酒率和酒精依賴的患病率均較高,長期大量飲酒者出現(xiàn)血脂代謝異常、肝功能異常等癥狀。醇脫氫酶3(ADH3)和醛脫氫酶2(ALDH2)基因多態(tài)性在嗜酒的形成中起一定作用,ADH3和ALDH2基因多態(tài)性可能對嗜酒的形成具有聯(lián)合作用。
[Abstract]:Objective to study the relationship between drinking behavior and genetic characteristics of Yugur men, and to provide scientific basis for the prevention and treatment of alcohol-related diseases. Methods 1) A self-assessment questionnaire was used to investigate the drinking status of 505 Yugur men in Sunan County. The influencing factors of drinking behavior were investigated by univariate and multivariate regression analysis. The alcohol-dependent patients were screened by using the Alcohol dependence Diagnostic scale (ADS). The distribution of blood lipids, liver function and renal function, and the polymorphism of alcohol dehydrogenase (ADH3) and acetaldehyde dehydrogenase (ALDH2) gene in spermatozoa dependent population and non-alcohol dependent population were compared. Polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP) was used to detect gene polymorphism. Results 1) the alcohol consumption rate of Yugur men was 56.83, and the prevalence of alcohol dependence was 9.90%. Logistic regression analysis showed that the independent correlation factors with drinking behavior included family income, marital status, daily smoking amount (positive correlation P 0.05, age, education, occupation). Health status (negative P0.05); factors with independent correlation with alcohol dependence included age, family income, marital status, daily smoking, annual alcohol consumption (positively correlated P0.05, educational level, occupation), There was a negative correlation between alcohol consumption and blood lipids, liver and kidney function, and the distribution of ALDH2O2 allele in health status (P 0.05), and the distribution of ALDH _ 2O _ 2 allele was negatively correlated with alcohol consumption. The allelic frequencies of ALDH2O1 and ALDH2O2 alleles in Yugur ethnic group were respectively: 76.82% and 33.18%, respectively. The frequencies of alleles of ALDH2O1 and ADH3O2 were: 87.87% and 16.13%, respectively. Conclusion the prevalence of alcohol consumption and alcohol dependence were higher in Yugur nationality men. The abnormal metabolism of blood lipids and liver function were found in a large number of alcohol drinkers for a long time. Alcohol dehydrogenase (ADH3) and aldehyde dehydrogenase (ALDH2) gene polymorphisms may play a role in the formation of alcoholism. ADH3 and ALDH2 gene polymorphisms may play a combined role in the formation of alcoholism.
