【摘要】：目的本實(shí)驗(yàn)通過(guò)溫腎補(bǔ)虛方對(duì)甲減大鼠的血脂及TSHR表達(dá)的影響,探討中醫(yī)藥治療甲減引起血脂異常的機(jī)制及方法。方法雄性SD大鼠隨機(jī)分為空白組、造模組,造模組予0.05%丙硫氧嘧啶(PTU)蒸餾水。造模成功后,造模組隨機(jī)分為模型組、西藥組、中低劑量組、中藥中劑量組、中藥膏劑量組,予相應(yīng)藥物,給藥時(shí)間4周。免疫組化法檢測(cè)TSHR的表達(dá);ELISA法檢測(cè)CHOL、LDL、TSH、T3、T4、FT3、FT4。結(jié)果造模后,大鼠血清CHOL、LDL、TSH、TSHR升高(P0.01),T3、T4、FT3、FT4降低(P0.01)。治療后,與模型組相比:中藥組、西藥組T3、T4、FT3、FT4升高顯著(P0.01);中藥組CHOL、LDL顯著下降(P0.05);中藥組及西藥組TSH極顯著下降(P0.01);中藥組TSHR水平顯著下降(P0.01)。結(jié)論溫腎補(bǔ)虛方能改善甲減大鼠CHOL、LDL、TSHR的含量。中藥調(diào)節(jié)CHOL、LDL的機(jī)制可能與降低TSHR的表達(dá)水平相關(guān)。
[Abstract]:Objective to explore the mechanism and methods of treating hypothyroidism with traditional Chinese medicine (TCM) through the effect of Wenshen Bushu recipe on blood lipid and TSHR expression in hypothyroidism rats. Methods male Sprague-Dawley rats were randomly divided into blank group. The model group was treated with 0.05% propylthiouracil (PTU) distilled water. After the establishment of the model, the model group was randomly divided into three groups: model group, western medicine group, middle and low dose group, middle dose group and ointment group, and the corresponding drugs were given for 4 weeks. The expression of TSHR was detected by immunohistochemical method. Results after the establishment of the model, the serum level of TSHHR of CHOLL LDL was increased, and the level of T3, T4, FT3, FT4 and P0.01 was decreased. After treatment, compared with the model group, the T3T4FT3FT4 in the western medicine group increased significantly (P 0.01); in the traditional Chinese medicine group, the Choll LDL decreased significantly (P 0.05); the TSH in the Chinese medicine group and the western medicine group decreased significantly (P 0.01); and the TSHR level in the Chinese medicine group decreased significantly (P 0.01). Conclusion Wenshen Bu deficiency prescription can improve the content of LDL TSHR in hypothyroidism rats. The mechanism of Chinese medicine regulating CHOLLL-LDL may be related to the decrease of TSHR expression level.
