【摘要】：目的分析白細(xì)胞介素2(IL-2)或IL-15協(xié)同結(jié)核分枝桿菌抗原(MTB-Ag)誘導(dǎo)人外周血單個(gè)核細(xì)胞(PBMC)擴(kuò)增細(xì)胞的生物學(xué)特性。方法采用MTB-Ag聯(lián)合IL-2或MTB-Ag聯(lián)合IL-15體外分別刺激正常人PBMC,培養(yǎng)至第12天時(shí),經(jīng)流式細(xì)胞術(shù)檢測擴(kuò)增細(xì)胞中γδT細(xì)胞、自然殺傷(NK)細(xì)胞和CD4+T細(xì)胞所占的比例,以及分析3種淋巴細(xì)胞擴(kuò)增的絕對數(shù)量。同時(shí)用MTT法檢測兩組擴(kuò)增細(xì)胞對腫瘤細(xì)胞的殺傷活性。結(jié)果與MTB-Ag聯(lián)合IL-2刺激組相比,MTB-Ag聯(lián)合IL-15刺激組擴(kuò)增細(xì)胞中γδT細(xì)胞所占比例明顯高于前者,NK細(xì)胞所占比例明顯低于前者;而兩組擴(kuò)增細(xì)胞中CD4~+T細(xì)胞所占比例并無顯著性差異。與2個(gè)刺激組中3種淋巴細(xì)胞數(shù)量的分析結(jié)果一致。并且MTB-Ag聯(lián)合IL-15刺激組擴(kuò)增細(xì)胞對不同腫瘤細(xì)胞均具有顯著的殺傷活性,但與MTB-Ag聯(lián)合IL-2刺激組相比,并無顯著性差異。結(jié)論 IL-15協(xié)同MTB-Ag可高效誘導(dǎo)人PBMC產(chǎn)生以γδT細(xì)胞增殖為主的效應(yīng)細(xì)胞,該擴(kuò)增細(xì)胞在體外對腫瘤細(xì)胞具有顯著的殺傷活性。
[Abstract]:Objective to investigate the biological characteristics of human peripheral blood mononuclear cells (PBMCs) induced by interleukin-2 (IL-2) or IL-15 in combination with Mycobacterium tuberculosis antigen (MTB-Ag). Methods normal PBMCs were stimulated by MTB-Ag combined with IL-2 or MTB-Ag combined with IL-15 in vitro. The percentage of 緯 未 T cells, natural killer NK cells and CD4 T cells were detected by flow cytometry at the 12th day of culture. The absolute number of lymphocyte amplification was analyzed. At the same time, MTT assay was used to detect the cytotoxicity of expanded cells to tumor cells. Results the percentage of 緯 未 T cells in the expanded cells of MTB-Ag + IL-15 group was significantly higher than that of the former group, but there was no significant difference between the two groups in the percentage of CD4T cells. The results were consistent with the results of the analysis of three kinds of lymphocytes in the two stimulation groups. The expanded cells of MTB-Ag combined with IL-15 had significant cytotoxicity to different tumor cells, but there was no significant difference compared with MTB-Ag combined with IL-2. Conclusion IL-15 combined with MTB-Ag can effectively induce the proliferation of 緯 未 T cells in human PBMC, and the expanded cells have significant cytotoxicity to tumor cells in vitro.
