脂肪干細胞成脂分化的分子機制和信號通路

【摘要】:背景:對脂肪干細胞成脂分化的研究不僅可探索肥胖的過程和機制,還可構建組織工程脂肪,為軟組織缺損的重建提供新的思路。目的:總結脂肪干細胞成脂分化的過程、分子機制和信號通路,討論miRNA對脂肪干細胞成脂分化的影響及脂肪干細胞成脂和成骨分化的關系。方法:應用計算機檢索Pub Med數(shù)據(jù)庫、Sino Med數(shù)據(jù)庫中有關脂肪干細胞分化的文獻,檢索詞為"adipose stem cell,adipose-derived stem cell,adipose-derived mesenchymal stem cell,differentiation;脂肪干細胞,分化",最終選取代表性文獻共27篇。結果與結論:在脂肪干細胞成脂分化過程中,PPARγ/MAPK、PI3K/Akt、Wnt/β-Catenin、cA MP、Notch等信號通路發(fā)揮了關鍵作用,但其相關基因和蛋白、信號通路和分子機制仍不完全清楚。miR NA對脂肪干細胞的成脂分化有重要影響,miR-24、miR-148a、miR-302等可促進脂肪干細胞的成脂分化,miR-22、miR-27等可抑制脂肪干細胞的成脂分化。脂肪干細胞的成骨和成脂分化關系密切,部分生長因子和激素在促進脂肪干細胞成脂分化的同時抑制其成骨分化,部分作用則相反。綜合利用各種細胞因子和支架模擬體內微環(huán)境,促進脂肪干細胞的增殖和成脂分化,構建組織工程脂肪,是未來的研究方向。
[Abstract]:Background: the study of adipogenic differentiation of adipose stem cells can not only explore the process and mechanism of obesity but also construct tissue engineered fat and provide a new idea for reconstruction of soft tissue defects. Aim: to summarize the process of adipogenic differentiation, molecular mechanism and signal pathway of adipose stem cells, and discuss the effect of miRNA on adipose stem cell adipogenic differentiation and the relationship between adipose stem cell adipogenesis and osteogenic differentiation. Methods: the literatures on adipose stem cell differentiation in Pub Med database Sino Med were searched by computer. The key words were "adipose stem cell adipose-derived stem adipose-derived mesenchymal stem cell differentiation; adipo-stem cell differentiation, differentiation". Results & conclusion: PPAR 緯 -MAPK / PI3K / 尾 -Catenin C A MPN Notch signaling pathway plays a key role in adipogenic differentiation of adipose stem cells, but its related genes and proteins play a key role in adipose stem cell adipogenic differentiation. The signaling pathway and molecular mechanism are still unclear. MIRNA plays an important role in adipogenic differentiation of adipose stem cells. MiR-24 miR-148amiR-302 can promote adipogenic differentiation. MiR-22miR-27 can inhibit adipogenic differentiation of adipose stem cells. The osteogenesis and adipogenic differentiation of adipose stem cells were closely related. Some growth factors and hormones inhibited adipose stem cells osteogenic differentiation while promoting adipose stem cell adipogenic differentiation. Comprehensive use of various cytokines and scaffolds to simulate the microenvironment, promote the proliferation and adipogenic differentiation of adipose stem cells, and construct tissue engineered fat, is the future research direction.
