維生素D3對自然流產(chǎn)模型小鼠胚胎丟失率及外周血免疫細(xì)胞的影響

【摘要】:目的探討維生素D3(VitD_3)對自然流產(chǎn)模型小鼠胚胎丟失率及外周血免疫細(xì)胞的影響。方法采用CBA/J×BALB/c建立正常妊娠小鼠模型,采用CBA/J×DBA/2建立自然流產(chǎn)小鼠模型,實驗動物分5組:正常組、自然流產(chǎn)組、VitD_3低劑量組、中劑量組和高劑量組,每組15只。VitD_3各組自妊娠第1天開始腹腔注射VitD_3溶液,單次VitD_3給藥劑量分別為1μg、4μg、16μg。記錄和觀察各組小鼠胚胎丟失情況,檢測各組血清調(diào)節(jié)性T細(xì)胞(Treg)細(xì)胞因子:白細(xì)胞介素-10(IL-10),輔助性T細(xì)胞17(Th17)細(xì)胞因子:白細(xì)胞介素-17A(IL-17A)以及各組外周血Treg、Th17。結(jié)果與正常組相比,自然流產(chǎn)組胚胎丟失率顯著升高(χ~2=16.045,P0.05);與自然流產(chǎn)組比較,VitD_3低劑量組、中劑量組和高劑量組胚胎丟失率顯著降低(χ~2=5.881、13.704、15.663,P0.05)。與正常組相比,自然流產(chǎn)組Treg、IL-10、Treg/Th17、IL-10/IL-17A顯著降低,Th17、IL-17A顯著升高,差異均有統(tǒng)計學(xué)意義(P0.05);與自然流產(chǎn)組比較,VitD_3低劑量組、中劑量組和高劑量組Treg、IL-10、Treg/Th17、IL-10/IL-17A顯著升高,Th17、IL-17A顯著降低,差異均有統(tǒng)計學(xué)意義(P0.05)。結(jié)論 VitD_3能夠降低自然流產(chǎn)模型小鼠胚胎丟失率,可能與調(diào)節(jié)Treg/Th17平衡有關(guān)。
[Abstract]:Objective to investigate the effect of Vitamin D _ (3) Vitamin D _ (3) on embryo loss and peripheral blood immune cells in spontaneous abortion mice. Methods normal pregnant mice were established by CBA/J 脳 BALB/c and spontaneous abortion by CBA/J 脳 DBA/2. The experimental animals were divided into 5 groups: normal group, spontaneous abortion group, low dose group, middle dose group and high dose group. VitD_3 solution was injected intraperitoneally from the first day of gestation to 15 rats in each group. The dose of VitD_3 was 1 渭 g / g 4 渭 g / g and 16 渭 g / g respectively. The embryo loss in each group was recorded and observed, and the cytokines of serum regulatory T cell (T cell), interleukin-10 (IL-10), helper T cell (17-Th17), interleukin-17A-1 (IL-17A) and peripheral blood Treg-Th17 (Th17) were detected in each group. Results compared with the normal group, the embryo loss rate in the spontaneous abortion group was significantly higher (蠂 ~ 2 / 216.045 / P _ (0.05), and the embryo loss rate in the medium and high dose groups was significantly lower than that in the low dose group (蠂 ~ 25.881) and the high dose group (蠂 ~ 2 / 25.881 ~ 13.704 ~ 15.663P0.05). Compared with the normal group, the Treg-IL-10 / Th17 / IL-17A level in the spontaneous abortion group was significantly lower than that in the control group (P 0.05), and the TregIL-10 Treg-10 Treg-10 / 10 / IL-17A in the low dose group, the middle dose group and the high dose group were significantly higher than that in the normal group, and the difference was statistically significant (P 0.05). Conclusion VitD_3 can reduce the rate of embryo loss in spontaneous abortion mice, which may be related to the regulation of Treg/Th17 balance.
