基于斑馬魚模型的26種常見骨傷科中藥材的毒性篩選

【摘要】:目的利用斑馬魚毒性模型評價26種常見傷科中藥材的安全性。方法將受精后1d(1dpf)的健康斑馬魚胚胎置于不同濃度藥味水煎液中,觀察記錄給藥后1~5d的死亡情況,顯微檢視3dpf或以后斑馬魚幼魚的形態(tài),同時設置0.4%DMSO溶媒對照組。用SPSS16.0計算不同藥味水煎液在6dpf的魚半數(shù)致死濃度LC_(50)。結果 13種藥味(續(xù)斷、知母、乳香加沒藥、補骨脂、獨活、牛膝、制何首烏、蛇床子、木香、土鱉蟲、淫羊藿和五加皮)水煎液致斑馬幼魚臟器形態(tài)明顯改變,毒性主要表現(xiàn)為卵黃囊腫大、變形、變黑,心包水腫、出血等,且它們的魚LC_(50)值較低(16.9~689.7μg/mL)。其余13種藥味(鹿銜草、千年健、蘇木、巴戟天、龜甲、骨碎補、丹參、狗脊、桑寄生、雞血藤、熟地黃、女貞子、杜仲),未見魚臟器毒性,且致魚死亡濃度高(在2 000μg/mL以上),較安全。結論用斑馬魚模型成功的篩選了26種常見骨傷科中藥的毒性,為骨傷科中藥臨床合理應用提供參考。斑馬魚模型具有簡單、高效、實時動態(tài)的優(yōu)勢,使中藥在體毒性規(guī)模化篩選成為可能。
[Abstract]:Objective to evaluate the safety of 26 common injured Chinese medicinal materials by zebrafish toxicity model. Methods the healthy zebrafish embryos of 1 day after fertilization were placed in different concentrations of medicinal flavor decoction. The death of juvenile zebrafish was observed and recorded at 1 ~ 5 days after administration. The morphology of juvenile zebrafish (3dpf) or the later zebrafish was examined microscopically, and the control group was set up with 0.4%DMSO solvent at the same time. SPSS16.0 was used to calculate the LD50 of fish with different medicinal flavors in 6dpf. Results 13 kinds of medicinal flavors (Dipsacus, Anemarrhena, frankincense plus myrrh, psoralen, Radix angelicae, Radix Polygoni multiflorum, Cnidium Cnidii, Radix Cynthopsis, hillbilly insect, Epimedium and Cortex Acanthopsis) were significantly changed in the organs of juvenile zebra fish. The main toxic manifestations were yolk sac swelling, deformation, blackening, pericardial edema, hemorrhage, etc. The remaining 13 kinds of medicinal flavors (Herba Deer, Chienjian, Sappa, Morinda officinalis, tortoise shell, shredder, salvia miltiorrhiza, dog ridge, mulberry parasitism, chicken blood rattan, cooked Rehmannia, Ligustrum lucidum, Eucommia ulmoides, no toxicity to fish viscera), It was safe to cause fish death (above 2 000 渭 g/mL). Conclusion the toxicity of 26 kinds of common orthopedic Chinese medicine was successfully screened by zebrafish model, which provides a reference for the rational clinical application of orthopedic Chinese medicine. Zebrafish model has the advantages of simple, efficient and real-time dynamic, which makes it possible to screen in vivo toxicity of traditional Chinese medicine on a large scale.
