【摘要】:目的:探討骨髓間充質(zhì)干細(xì)胞(bone marrow mesenchymal stem cells,BMSCs)不同途徑移植對(duì)四氯化碳誘發(fā)的肝硬化模型大鼠的治療效果差異。 方法:將清潔級(jí)Sprague-Dawley大鼠分為4組,A組(正常對(duì)照組)、B組(空白模型組)、C組(骨髓間充質(zhì)干細(xì)胞肝動(dòng)脈移植組)、D組(骨髓間充質(zhì)干細(xì)胞門靜脈移植組)。用四氯化碳分析純與橄欖油混合液皮下注射和酒精喂養(yǎng)的方法建立肝硬化大鼠模型。取健康SD大鼠骨髓,使用Percoll液離心分離,利用全骨髓貼壁的方法原代培養(yǎng)骨髓間充質(zhì)干細(xì)胞,流式細(xì)胞儀檢測(cè)細(xì)胞表面抗原,然后通過2種途徑分別進(jìn)行移植。術(shù)后2周檢測(cè)血清白蛋白(ALB)及谷丙轉(zhuǎn)氨酶(ALT),ELISA法檢測(cè)包括血清透明質(zhì)酸酶(HA)、血清Ⅳ型膠原(CⅣ)、血清Ⅲ型前膠原(PcⅢ)、層粘蛋白(LN)等肝纖維化四項(xiàng)指標(biāo),,取各組大鼠肝組織切片行HE和Masson染色,觀察肝硬化程度 結(jié)果:通過2種途徑移植BMSCs到肝硬化模型大鼠體內(nèi),2周后發(fā)現(xiàn)接受移植組模型大鼠的肝臟組織鏡下表現(xiàn)以及肝功能均得到明顯改善,血清白蛋白較肝硬化未移植組顯著升高(P0.05),谷丙轉(zhuǎn)氨酶較肝硬化未移植組顯著降低(P0.05),肝纖維化四項(xiàng)較肝硬化未移植組顯著減輕(P0.05)。2移植途徑不同組的大鼠相關(guān)指標(biāo)用SPSS軟件對(duì)比分析,肝纖維化組織學(xué)半定量計(jì)分對(duì)比無統(tǒng)計(jì)學(xué)差異(P0.05),肝功能和肝纖維化四項(xiàng)等生化指標(biāo)則肝動(dòng)脈途徑移植組要略好于門靜脈途徑移植組(P0.05)。 結(jié)論:肝硬化模型大鼠接受骨髓間充質(zhì)干細(xì)胞移植,移植后2周肝功能改善,肝纖維化程度降低,肝動(dòng)脈途徑移植組的指標(biāo)要略好于門靜脈途徑移植組,結(jié)合臨床,應(yīng)作為首選途徑。
[Abstract]:Aim: to investigate the therapeutic effects of bone marrow mesenchymal stem cells BMSCs transplantation on carbon tetrachloride induced cirrhotic rats. Methods: clean grade Sprague-Dawley rats were divided into 4 groups: group A (normal control group, group B) (group C, blank model group), group D (portal vein transplantation of bone marrow mesenchymal stem cells) (group D), group D (portal vein transplantation of bone marrow mesenchymal stem cells). The liver cirrhosis rat model was established by carbon tetrachloride (CCL 4) analysis, subcutaneous injection of pure olive oil mixture and alcohol feeding. Bone marrow of healthy SD rats was isolated by centrifugation with Percoll solution. Bone marrow mesenchymal stem cells were primary cultured by whole bone marrow adherent method. Flow cytometry was used to detect cell surface antigen. Two weeks after operation, serum albumin (ALB) and alanine aminotransferase (alt) were detected by Elisa, including serum hyaluronidase (HA), serum type 鈪
