【摘要】：目的探討重樓總皂甙對(duì)小鼠哮喘模型的保護(hù)作用及可能機(jī)制。方法將24只BALB/c小鼠隨機(jī)分為空白對(duì)照組,模型組,重樓總皂甙低劑量組(LRTPS)和重樓總皂甙高劑量組(HRTPS),每組6只。除空白對(duì)照組小鼠外,其余各組采用卵蛋白致敏建立支氣管哮喘小鼠模型,治療組在造模的同時(shí)灌胃給予不同劑量的重樓總皂甙治療。通過肺組織病理切片、酶聯(lián)免疫反應(yīng)(ELISA)法檢測(cè)肺泡灌洗液中的白細(xì)胞介素(IL)-4和干擾素(IFN)-γ水平,Q-PCR檢測(cè)肺組織IL-4和IFN-γmRNA表達(dá)水平。結(jié)果空白對(duì)照組小鼠肺組織支氣管無(wú)炎性細(xì)胞浸潤(rùn),氣道無(wú)明顯黏液分泌,模型組支氣管和血管周圍炎性細(xì)胞浸潤(rùn)明顯,黏膜及黏膜下層水腫明顯。治療組支氣管和血管周圍炎性細(xì)胞浸潤(rùn)明顯減少,肺泡結(jié)構(gòu)清晰,肺泡腔和支氣管腔無(wú)滲出物。哮喘組小鼠的肺泡灌洗液IL-4水平和肺組織IL-4 mRNA表達(dá)顯著高于對(duì)照組;治療組顯著低于哮喘組(P0.05)。哮喘組小鼠的肺泡灌洗液IFN-γ水平和肺組織IFN-γmRNA表達(dá)顯著低于對(duì)照組,治療組顯著高于哮喘組(P0.05)。結(jié)論重樓總皂甙能明顯減輕哮喘氣道炎性反應(yīng),其機(jī)制可能與恢復(fù)局部Th1/Th2細(xì)胞因子平衡有關(guān)。
[Abstract]:Objective to investigate the protective effect and possible mechanism of total saponins in mice with asthma. Methods Twenty-four BALB/c mice were randomly divided into control group, model group, low dose group and high dose group with 6 rats in each group. In addition to the blank control group, the other groups were sensitized with ovalbumin to establish the model of bronchial asthma. The treatment group was treated with different doses of total saponins in the stomach at the same time. The levels of IL-4 and IFN- 緯 in alveolar lavage fluid were detected by enzyme-linked immunosorbent assay (Elisa). Q-PCR was used to detect the expression of IL-4 and IFN- 緯 mRNA in lung tissue. Results there were no inflammatory cells infiltration and mucus secretion in the bronchus of the blank control group. In the model group, the infiltration of inflammatory cells in the bronchus and perivascular was obvious, and the edema of mucosa and submucous membrane was obvious in the model group. In the treatment group, the infiltration of inflammatory cells in the bronchi and blood vessels was decreased, the alveolar structure was clear, and there was no exudate in the alveolar and bronchial lumen. The level of IL-4 in alveolar lavage fluid and the expression of IL-4 mRNA in lung tissue in asthmatic group were significantly higher than those in control group, while those in treatment group were significantly lower than those in asthma group. The level of IFN- 緯 in alveolar lavage fluid and the expression of IFN- 緯 mRNA in lung tissue in asthma group were significantly lower than those in control group, and the level of IFN- 緯 in treatment group was significantly higher than that in asthma group. Conclusion the total saponins can significantly reduce the airway inflammatory response of asthma, and the mechanism may be related to the restoration of local Th1/Th2 cytokine balance.
