遠志石菖蒲藥對對注意缺陷多動障礙模型大鼠夜周期行為學的影響

【摘要】:目的探討遠志-石菖蒲藥對治療注意缺陷多動障礙(ADHD)的可能作用機制。方法將30只自發(fā)性高血壓(SHR)大鼠隨機分為模型組、陽性對照組、遠志-石菖蒲組,每組10只,另設WKY大鼠10只為對照組。遠志-石菖蒲組給予遠志-石菖蒲混懸液4.76 mg/(kg·d)灌胃;陽性對照組給予利他林混懸液2mg/(kg·d)灌胃;正常組和模型組均以0.5%羧甲基纖維素鈉溶液2 ml/(kg·d)灌胃,在大鼠夜周期9:30~10:30、15:30~16:30分兩次灌胃,連續(xù)給藥4周。分別于給藥前、給藥4周及停藥2周時,采用開場實驗評價大鼠自發(fā)性行為;給藥4周采用高架十字迷宮、新物體識別實驗評估大鼠沖動行為及學習記憶功能。停藥2周時檢測前額葉皮質中多巴胺(DA)、多巴胺轉運體(DAT)水平。結果給藥4周遠志-石菖蒲組大鼠在開場實驗中的總運動距離及直立次數、進入高架十字迷宮開臂次數百分比和開臂滯留時間百分比均低于模型組(P0.05),而在新物體識別實驗對新物體的偏好指數則高于模型組(P0.05);停藥2周遠志-石菖蒲組大鼠總運動距離及直立次數仍少于模型組,前額葉皮質中DA水平較模型組明顯升高(P0.05),而DAT含量與模型組間差異無統(tǒng)計學意義(P0.05)。結論遠志-石菖蒲藥對可提高ADHD小鼠前額葉皮質中DA含量,可能是其治療ADHD的作用機制之一。
[Abstract]:Objective to explore the possible mechanism of Radix Polygali-Acorus calamus on ADHD in the treatment of attention deficit hyperactivity disorder (ADHD). Methods Thirty spontaneously hypertensive SHRs were randomly divided into three groups: model group, positive control group, Rhizoma Polygala and Acorus calamus group with 10 rats in each group and 10 WKY rats as control group. The control group was given Ritalin suspension (2mg/(kg d), the normal group and the model group were given 0.5% sodium carboxymethyl cellulose solution for 2 ml/(kg d, and the rats were perfused twice at 9: 30: 30: 3015: 3016: 30 during the night period of 9: 30: 30: 3015: 30: 30: 30, respectively, and the rats in the model group were treated with 0.5% sodium carboxymethyl cellulose solution for 2 ml/(kg d. The drug was given continuously for 4 weeks. The spontaneous behavior of rats was evaluated by open field experiment before administration, 4 weeks before administration and 2 weeks after withdrawal, and by using elevated cross maze at 4 weeks. The impulsive behavior and learning and memory function of rats were evaluated by new object recognition experiment. At 2 weeks after withdrawal, the levels of dopamine cinnamon and dopamine transporter were detected in the prefrontal cortex. Results the total distance of movement and the number of times of standing up in the open field experiment of the rats in the group of Polygala acuminata and Acorus calamus were given for 4 weeks. The percentage of open arm times and open arm retention time in the elevated cross maze was lower than that in the model group, but the preference index of the new object recognition experiment was higher than that of the model group. The total movement distance and the number of upright times were still less than those of the model group. The DA level in the prefrontal cortex was significantly higher than that in the model group (P 0.05), but there was no significant difference between the DAT content and the model group (P 0.05). Conclusion Radix Polygala and Acorus calamus can increase DA content in prefrontal cortex of ADHD mice, which may be one of the mechanisms of its therapeutic effect on ADHD.
