細(xì)胞外ATP對(duì)失神經(jīng)大鼠腓腸肌細(xì)胞中FoXO3a通路的影響

【摘要】:目的:探討細(xì)胞外ATP對(duì)失神經(jīng)大鼠腓腸肌細(xì)胞中FoXO3a/MAFbX通路的影響。 方法:雌性成年Wistar大鼠54只隨機(jī)分三組,ATP治療組和失神經(jīng)對(duì)照組在右側(cè)梨狀肌下緣切斷坐骨神經(jīng),制作失神經(jīng)動(dòng)物模型。術(shù)后ATP治療組于右側(cè)腓腸肌內(nèi)注射ATP 0. 1mg/ d,左側(cè)腓腸肌內(nèi)注射等量生理鹽水;失神經(jīng)對(duì)照組雙下肢注射等量的生理鹽水,正常對(duì)照組(0天)不做任何處理。在術(shù)后0,2,7,14,,28天分別處死一組大鼠取其兩側(cè)腓腸肌,稱肌濕重,計(jì)算肌失重比(右側(cè)"左側(cè)),采用年實(shí)時(shí)熒光定量PCR和Western Blotting檢測(cè)大鼠腓腸肌FoxO3a、MAFbx的mRNA和蛋白表達(dá)水平以及253位磷酸化的FoxO3a(p-FoxO3aS253)蛋白表達(dá)。 結(jié)果:ATP治療組肌濕重比高于失神經(jīng)對(duì)照組,差異具有統(tǒng)計(jì)學(xué)意義(p<0.01)。ATP治療組FoxO3amRNA和蛋白表達(dá)略低于失神經(jīng)照組,差異無(wú)統(tǒng)計(jì)學(xué)意義(p>0.05)。MAFbxmRNA和蛋白表達(dá)低于失神經(jīng)對(duì)照組,差異有統(tǒng)計(jì)學(xué)意義(p<0.05)。p-FoxO3a蛋白表達(dá)量較對(duì)照組明顯增加,不同時(shí)段差異均具有統(tǒng)計(jì)學(xué)意義(p<0.05)。 結(jié)論:細(xì)胞外ATP可能通過(guò)磷酸化FoxO3a進(jìn)而抑制MAFbX蛋白的表達(dá)來(lái)延緩去神經(jīng)性大鼠肌萎縮。
[Abstract]:Aim: to investigate the effect of extracellular ATP on FoXO3a/MAFbX pathway in gastrocnemius muscle cells of denervated rats. Methods: Fifty-four female adult Wistar rats were randomly divided into three groups: the ATP-treated group and the denervated control group. The sciatic nerve was cut off at the lower edge of the right piriformis muscle to make the denervated animal model. ATP was injected into the right gastrocnemius muscle in the ATP group after operation. 1mg/ d, the left gastrocnemius muscle was injected with the same amount of saline in the left gastrocnemius muscle and the denervated control group was injected with the same amount of saline in the lower extremities of the denervated group, and the normal control group was given 0 days The gastrocnemius muscles were taken from both sides of the gastrocnemius muscle and weighed wet on day 1428 after operation. The muscle weight loss ratio (right) was calculated. The expression of mRNA and protein in the gastrocnemius muscle FoxO3a, MAFbx and phosphorylated FoxO3ap-FoxO3aS253 protein were detected by annual real-time quantitative PCR and Western Blotting. Results the wet weight ratio of muscle in the treatment group was higher than that in the denervated control group, and the difference was statistically significant (P < 0.05). The expression of FoxO3amRNA and protein in the 0.01).ATP group was slightly lower than that in the denervated group. There was no significant difference in the expression of 0.05).MAFbxmRNA and protein between the two groups. The difference was statistically significant (P < 0.05). The expression of 0.05).p-FoxO3a protein was significantly higher than that of the control group. Conclusion: extracellular ATP may delay denervated rat muscle atrophy by phosphorylated FoxO3a and inhibit the expression of MAFbX protein.
