不同代次人臍帶間充質(zhì)干細(xì)胞的生物學(xué)特性及分化

【摘要】:背景:人臍帶間充質(zhì)干細(xì)胞作為組織工程及再生醫(yī)學(xué)領(lǐng)域的新興種子細(xì)胞,其應(yīng)用價(jià)值有賴于能否在長期體外傳代擴(kuò)增后仍保持穩(wěn)定生物學(xué)特性和多向分化能力,需要進(jìn)一步的實(shí)驗(yàn)研究。目的:研究不同培養(yǎng)代次人臍帶間充質(zhì)干細(xì)胞重要生物學(xué)屬性的差異。方法:采用組織塊法分離培養(yǎng)人臍帶間充質(zhì)干細(xì)胞,分別采用細(xì)胞計(jì)數(shù)法與MTT法檢測第3-6代細(xì)胞增殖活性;流式細(xì)胞儀檢測第1-9代細(xì)胞表面標(biāo)志物;細(xì)胞染色法分析第3-6代細(xì)胞核型。取第3代人臍帶間充質(zhì)干細(xì)胞,檢測其分化為神經(jīng)細(xì)胞、成骨細(xì)胞及肝樣細(xì)胞的能力。結(jié)果與結(jié)論:(1)細(xì)胞計(jì)數(shù)法與MTT方法檢測結(jié)果一致,第3-6代臍帶間充質(zhì)干細(xì)胞的增殖曲線均呈"S"型,在一二天時(shí)處于緩慢增長期,2-4 d時(shí)處于對數(shù)增長期,四五天時(shí)處于平臺期,五六天時(shí)處于衰退期;(2)第1-9代臍帶間充質(zhì)干細(xì)胞表面高表達(dá)CD90、CD44、CD73,不或低表達(dá)CD11b、CD18、CD34、CD43及HLA-DR;(3)第3-6代臍帶間充質(zhì)干細(xì)胞核型均為正常人體核型;(4)臍帶間充質(zhì)干細(xì)胞可分化為神經(jīng)細(xì)胞、成骨細(xì)胞及肝樣細(xì)胞;(5)結(jié)果表明,人臍帶間充質(zhì)干細(xì)胞具有高度增殖活性和多向分化特性。
[Abstract]:Background: as a new seed cell in tissue engineering and regenerative medicine, the application value of human umbilical cord mesenchymal stem cells depends on the ability to maintain stable biological characteristics and multidirectional differentiation after long-term passage and amplification in vitro. Further experimental research is needed. Objective: to study the difference of important biological properties of human umbilical cord mesenchymal stem cells in different generations. Methods: human umbilical cord mesenchymal stem cells were isolated and cultured by tissue mass method. The proliferative activity of passage 3-6 cells was detected by cell count method and MTT method, and cell surface markers of passage 1-9 were detected by flow cytometry. The nuclear types of passage 3-6 were analyzed by cell staining. The third generation of human umbilical cord mesenchymal stem cells were taken and their ability to differentiate into neural cells, osteoblasts and hepatoid cells was tested. The results were consistent with those of MTT method. The proliferation curve of umbilical cord mesenchymal stem cells of passage 3 to 6 was "S" type, and it was in logarithmic growth phase at 2 ~ 4 days after slow growth on day 1 and 2, and at platform stage on day 45. During the 5th and 6th days, the mesenchymal stem cells of the umbilical cord expressed CD90, CD4, CD73, CD11, CD18, CD34, CD43 and HLA-DR3, respectively. The nuclear types of the mesenchymal stem cells of the 3rd and 6th generations of cord were all normal human karyotype and the mesenchymal stem cells could differentiate into neural cells, and the mesenchymal stem cells of the umbilical cord could differentiate into neural cells, and the mesenchymal stem cells of the third and sixth generations could differentiate into neural cells. The results of osteoblasts and hepatoid cells showed that human umbilical cord mesenchymal stem cells had high proliferative activity and multidirectional differentiation.
