【摘要】：目的針對NKT細胞培養(yǎng)擴增和培養(yǎng)過程中遇到的過早凋亡現(xiàn)象,在凋亡信號層面進行初步的描述和探討。為今后的NKT相關(guān)研發(fā)提供數(shù)據(jù)參考。方法自健康人濃縮白細胞制品(白膜)分離有外周血單個核細胞(peripheral blood mononuclear cells PBMNCs),利用其中的抗原遞呈細胞,以α-Gal Cer的刺激NKT細胞克隆產(chǎn)生,記錄3-4周NKT的表型變化,和Caspase-3酶活化及細胞凋亡狀況。結(jié)果刺激7 d后,NKT克隆開始產(chǎn)生,持續(xù)擴增3周左右后開始呈現(xiàn)凋亡狀態(tài),Caspase3酶在第2周即開始維持高活性狀態(tài)。人NKT細胞對α-Gal Cer的刺激的反應(yīng)具有很大的個體差異。結(jié)論 NKT細胞克隆可以再最初的3周內(nèi)如同常規(guī)T細胞克隆一樣生長,其后則會因α-GalCer刺激引發(fā)活化后凋亡。
[Abstract]:Objective to investigate the phenomenon of premature apoptosis in the process of NKT cell proliferation and culture. For the future NKT related research and development to provide data reference. Methods Peripheral blood mononuclear cells (PBMNCs) were isolated from the white membrane of healthy people. The antigen-presenting cells were used to produce NKT cells stimulated by 偽 -Gal Cer, and the phenotypic changes of NKT were recorded at 3-4 weeks. And Caspase-3 enzyme activation and apoptosis. Results after stimulation for 7 days, NKT clones began to produce, and then continued to amplify for about 3 weeks. The apoptotic state of Caspase3 enzyme began to maintain high activity at the second week. The response of human NKT cells to 偽 -Gal Cer is very different. Conclusion NKT cell clone can grow as conventional T cell clone in the first 3 weeks, and then apoptosis after activation induced by 偽 -GalCer stimulation.
