【摘要】：目的探討歸芪益元膏防治重離子(12C6+)束輻射肺腎旁效應(yīng)的作用機(jī)制,為臨床輻射損傷防護(hù)提供新的思路。方法健康雄性Wistar大鼠60只,隨機(jī)分為正常對照組(NCG組)、單純輻射組(SRG組)、歸芪益元膏組(GOG組),每組20只。灌胃給藥7d,12C6+束輻射右側(cè)肺部造模,造模后48h處死,取大鼠左肺、左腎及右腎組織,ELISAlike法檢測DNA甲基化率,HE染色觀察組織病理學(xué)改變,免疫組織化學(xué)法檢測Dnmt1、Dnmt3a、Dnmt3b的表達(dá)。結(jié)果與正常對照組比較,單純輻射組大鼠DNA甲基化水平顯著降低(P0.01);左肺有炎癥,左右腎無異常;Dnmt1、Dnmt3a、Dnmt3b表達(dá)明顯升高(P0.01)。與單純輻射組比較,歸芪益元膏組大鼠DNA甲基化水平升高(P0.01);左肺炎癥減輕;Dnmt1、Dnmt3a、Dnmt3b表達(dá)降低(P0.01)。Dnmt1、Dnmt3a、Dnmt3b蛋白均在支氣管黏膜上皮細(xì)胞、腎小管上皮細(xì)胞的胞質(zhì)內(nèi)表達(dá),各組均可見;正常對照組呈淺褐色-褐色染色,為弱陽性表達(dá);單純輻射組呈褐色-棕色染色,為強(qiáng)陽性表達(dá);歸芪益元膏組呈淺褐色-棕褐色染色,為中等程度陽性表達(dá)。結(jié)論歸芪益元膏可減輕12C6+束輻射造成的肺腎旁效應(yīng),其機(jī)制可能與調(diào)控DNA甲基化水平有關(guān)。
[Abstract]:Objective to explore the mechanism of Guiqi Yiyuan ointment in preventing and treating lung and kidney side effects of heavy ion 12C6 beam radiation, and to provide a new idea for the protection of clinical radiation injury. Methods Sixty healthy male Wistar rats were randomly divided into normal control group (n = 20), radiation group (n = 20) and Guiqi Yiyuan ointment group (n = 20). The right lung model was induced by intragastric administration of 12C6 beam on the 7th day. The left lung, left kidney and right kidney were sacrificed 48 hours later. The DNA methylation rate and the histopathological changes were detected by ELISA-like method, and the expression of Dnmt1Dnmt3aDnmt3b was detected by immunohistochemistry. Results compared with the normal control group, the DNA methylation level in the radiation alone group was significantly lower than that in the control group, while there was inflammation in the left lung, and the expression of Dnmt3a1 + Dnmt3b in the left and right kidneys was significantly higher than that in the control group (P 0.01). Compared with the radiation group, the DNA methylation level of rats in the Guiqi Yiyuan ointment group was increased (P 0.01), and the expression of Dnmt3a Dnmt3b was reduced in the left lung inflammation group, and the expression of Dnmt3b protein in the epithelial cells of bronchial mucosa and renal tubules were all observed in all the groups, and the expression of Dnmt3b protein was found in the epithelial cells of bronchial mucosa and renal tubules. The normal control group showed light brown-brown staining with weak positive expression; the radiation group showed brown-brown staining with strong positive expression; the Guiqi Yiyuan ointment group showed light brown-tan staining with moderate positive expression. Conclusion Guiqi Yiyuan ointment can reduce the pulmonary and renal effects induced by 12C6 beam radiation, and its mechanism may be related to the regulation of DNA methylation level.
