【摘要】：目的制作一種簡單、方便,適合新生大鼠灌胃給藥的灌胃器。方法選取141只新生大鼠,按隨機數(shù)表法分4組,即對照組與低、中、高劑量組。采用新型灌胃器于生后48 h內(nèi)對每只新生大鼠灌予生理鹽水或二十二碳六烯酸(DHA)600、300、100 mg/kg,1次/d,0.01 m L/(g·次),共灌喂21 d,觀察不同劑量藥物對新生鼠的存活情況、體質(zhì)量變化及神經(jīng)發(fā)育的影響。結果 128只新生鼠存活,動物存活率90.78%。新生鼠出生體重(7.37±0.063)g。7日齡體重:對照組與低、中、高劑量組分別為(18.57±0.76)、(1.66±0.89)、(14.23±0.49)、(16.13±1.02)g;14日齡體質(zhì)量分別為(37.60±1.32)、(36.24±0.84)、(33.63±1.04)、(31.69±1.77)g;21日齡體質(zhì)量分別為(53.01±11.75)、(58.52±6.62)、(52.47±8.18)、(45.90±13.17)g。順利完成實驗。結論新型灌胃器對新生大鼠的口腔、食管黏膜的損傷小,給藥依從性好、給藥時間快速,新生大鼠存活率高,適合用于對大批量的新生大鼠進行長期、有效的灌胃,有利于順利完成實驗過程,為通過新生大鼠進行藥物、營養(yǎng)的相關研究提供了建制新生大鼠動物模型的良好的方法與依據(jù)。
[Abstract]:Objective to make a simple, convenient and suitable gavage apparatus for the fresh-born rats. Methods 141 newborn rats were selected and divided into 4 groups according to the random number table method. The control group and the low, middle and high dose group were given a new type of intragastric irrigator in 48 h after birth to each newborn rat with physiological salt water or twenty-two carbon six enoic acid (DHA) 600300100 mg/kg, 1 times /d, 0 .01 m L/ (G.) was fed a total of 21 d to observe the survival of the newborn rats, the changes of body mass and the effect of nerve development. Results 128 newborn rats survived, the survival rate of the newborn rats was 90.78%. birth weight (7.37 + 0.063) G.7 day old weight: the control group was (18.57 + 0.76) and (1.66 + 0.89) in the low, middle and high dose group, (14.23. 0.89), 14.23. (16.13 + 1.02) g, 14 days old body mass were (37.60 + 1.32), (36.24 + 0.84), (33.63 + 1.04) and (31.69 + 1.77), respectively. The mass of 21 days age body was respectively (0.84 + 37.60), and g. successfully completed the experiment. Conclusion the new type of gavage apparatus has little damage to the oral mucosa of newborn rats, and the compliance of the administration is good. The time of drug delivery is rapid and the survival rate of newborn rats is high. It is suitable for the long-term and effective gastric perfusion of large batch of neonatal rats, which is beneficial to the successful completion of the experimental process. The related research on nutrition provides a good method and basis for the establishment of the model of the new rat.
