【摘要】:背景:研究發(fā)現(xiàn)降鈣素基因相關(guān)肽能夠增強(qiáng)成骨細(xì)胞的生物學(xué)活性,實(shí)驗(yàn)擬進(jìn)一步檢測(cè)降鈣素基因相關(guān)肽對(duì)骨髓間充質(zhì)干細(xì)胞成骨分化的影響。目的:探討外源性降鈣素基因相關(guān)肽對(duì)去勢(shì)大鼠骨髓間充質(zhì)干細(xì)胞增殖、成骨能力的影響。方法:切除雌性SD大鼠雙側(cè)卵巢建立骨質(zhì)疏松模型,并通過(guò)micro-CT驗(yàn)證。從假手術(shù)和去勢(shì)大鼠股骨中提取骨髓間充質(zhì)干細(xì)胞,培養(yǎng)液中添加不同濃度降鈣素基因相關(guān)肽,MTT法檢測(cè)細(xì)胞增殖能力。對(duì)骨髓間充質(zhì)干細(xì)胞進(jìn)行成骨誘導(dǎo),在誘導(dǎo)液中添加不同濃度降鈣素基因相關(guān)肽,成骨誘導(dǎo)7 d,采用Real time RT-PCR和Western blot檢測(cè)成骨特異性蛋白R(shí)unx2,Osterix的表達(dá),成骨誘導(dǎo)21 d進(jìn)行茜素紅染色。結(jié)果與結(jié)論:(1)切除雌性大鼠雙側(cè)卵巢可成功建立骨質(zhì)疏松模型;(2)不同濃度的降鈣素基因相關(guān)肽可增強(qiáng)去勢(shì)大鼠骨髓間充質(zhì)干細(xì)胞的增殖及成骨分化能力,且具有劑量依賴性;(3)結(jié)果表明,降鈣素基因相關(guān)肽對(duì)去勢(shì)大鼠骨髓間充質(zhì)干細(xì)胞增殖及成骨分化能力有直接促進(jìn)作用,從而有利于骨形成。
[Abstract]:Background: calcitonin gene-related peptide (CGRP) has been found to enhance the biological activity of osteoblasts. The effect of calcitonin gene-related peptide on osteogenic differentiation of bone marrow mesenchymal stem cells was studied. Aim: to investigate the effects of exogenous calcitonin gene related peptide (CGRP) on proliferation and osteogenesis of bone marrow mesenchymal stem cells (MSCs) in ovariectomized rats. Methods: Osteoporosis model was established by excision of bilateral ovaries of female SD rats and verified by micro-CT. Bone marrow mesenchymal stem cells (BMSCs) were extracted from sham-operated and ovariectomized rats femur. The proliferation of MSCs was detected by MTT assay with different concentrations of calcitonin gene-related peptide. Bone marrow mesenchymal stem cells (BMSCs) were induced by osteogenesis. Calcitonin gene-related peptide (CGRP) was added to the induction fluid for 7 days. Real time RT-PCR and Western blot were used to detect the expression of the osteoblast specific protein Runx2Osterix. Alizarin red staining was performed on 21 days after osteogenesis induction. Results and conclusion Osteoporosis model could be established successfully by removing female rat bilateral ovaries. Different concentrations of calcitonin gene-related peptide could enhance the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells in ovariectomized rats. The results showed that calcitonin gene-related peptide could directly promote the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells in ovariectomized rats.
