胎鼠皮膚來源肥大細(xì)胞的分離培養(yǎng)和鑒定

【摘要】:目的分離并誘導(dǎo)培養(yǎng)胎鼠皮膚來源肥大細(xì)胞。方法分離14~16 d胎鼠皮膚細(xì)胞并加入重組小鼠白細(xì)胞介素3(IL-3)和重組小鼠干細(xì)胞因子共同誘導(dǎo)其中的肥大細(xì)胞分化成熟。細(xì)胞培養(yǎng)14 d后,經(jīng)密度梯度離心法得到成熟肥大細(xì)胞。采用甲苯胺藍(lán)染色或類胰蛋白酶免疫熒光細(xì)胞化學(xué)染色檢測(cè)判斷肥大細(xì)胞純度,流式細(xì)胞術(shù)檢測(cè)Ig E的高親和力受體(FcεRⅠ)和CD117進(jìn)一步鑒定肥大細(xì)胞純度。結(jié)果培養(yǎng)14 d并純化后,細(xì)胞經(jīng)甲苯胺藍(lán)及免疫熒光染色可見胞質(zhì)內(nèi)大量顆粒,FcεR I+CD117+細(xì)胞達(dá)97%。結(jié)論成功從胎鼠皮膚分離并獲得大量高純度成熟的肥大細(xì)胞。
[Abstract]:Objective to isolate and induce cultured mast cells from fetal rat skin. Methods the skin cells of fetal mice were isolated for 14 days and the differentiation and maturation of mast cells were induced by adding recombinant mouse interleukin-3 (IL-3) and recombinant mouse stem cell factor (RSCF). After cultured for 14 days, mature mast cells were obtained by density gradient centrifugation.
