應(yīng)用兩種不同建模方式構(gòu)建子癇前期SD大鼠動物模型的實(shí)驗(yàn)研究

【摘要】:目的:探討分析應(yīng)用兩種不同建模方式來構(gòu)建子癇前期SD大鼠的動物模型的特異性。方法:SD孕鼠隨機(jī)均分3組,空白對照組同前飼養(yǎng),合體滋養(yǎng)細(xì)胞微絨毛(STBM)組用STBM誘導(dǎo),STBM+亞硝基左旋精氨酸甲酯(L-NAME)組用STBM+L-NAME誘導(dǎo)。檢測蛋白尿(U-TP)、血壓(BP)、部分凝血活酶時(shí)間(APTT)、凝血酶原時(shí)間(PT)和D-二聚體(D-D),HE染色胎盤組織,Real time PCR和Western blot測胎盤組織可溶性血管內(nèi)皮生長因子受體-1(s Flt-1)、可溶性內(nèi)皮因子(SEng)、胎盤生長因子(PLGF)mRNA及蛋白表達(dá),滋養(yǎng)細(xì)胞分離、培養(yǎng)后行功能學(xué)實(shí)驗(yàn)(EDU測增殖、FCM測凋亡、Transwell測侵襲)。結(jié)果:U-TP、BP、APTT、PT及D-D在孕10天3組比較差異無統(tǒng)計(jì)學(xué)意義(P0.05),孕15天及20天U-TP、BP STBM+L-NAME組較STBM組顯著增加(P0.05),孕20天APTT、PT STBM+L-NAME組較STBM組顯著縮短,D-D顯著增加,空白對照組無明顯變化。STBM+L-NAME組HE染色見胎盤蛻膜帶水腫明顯,炎性細(xì)胞浸潤,基帶和迷路帶見滋養(yǎng)細(xì)胞增生。3組比較s Flt-1、SEng、PLGF mRNA及蛋白表達(dá)差異有統(tǒng)計(jì)學(xué)意義(P0.05)。STBM+L-NAME組s Flt-1、SEng mRNA及蛋白比STBM組明顯增高(P0.05),PLGF mRNA及蛋白明顯減低(P0.05)。功能學(xué)實(shí)驗(yàn)中3組細(xì)胞增殖和凋亡差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。STBM+L-NAME組滋養(yǎng)細(xì)胞增殖和侵襲最少,凋亡最多。與STBM組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:在構(gòu)建子癇前期動物模型中,STBM合用L-NAME誘導(dǎo)比單獨(dú)應(yīng)用STBM特異性更強(qiáng),為臨床子癇前期診療提供可靠模式研究依據(jù)。
[Abstract]:Aim: to investigate the specificity of two different modeling methods in constructing preeclampsia SD rat model. Methods Twenty SD pregnant rats were randomly divided into 3 groups. The control group was fed with STBM L-NAME. The syncytiotrophoblast microvillus chorionic chorionic chorionic microvilli (STBM) group was induced by STBM L-NAME. Detection of proteinuria (UTP), blood pressure (BP), partial thromboplastin time (APTTT), prothrombin time (PTT) and D- dimer (D-DX) HE staining of placental tissue Real time PCR and Western blot for the detection of soluble vascular endothelial growth factor receptor -1s Flt-1, soluble endothelial factor (et) The mRNA and protein expression of placental growth factor-PLGFN were observed. Cultured trophoblastic cells were separated and cultured. EDU was used to measure proliferation and FCM to detect apoptosis and Transwell to detect invasion. Results there was no significant difference in PT and D-D between the three groups on the 10th day of gestation. On the 15th and 20th day of gestation, the levels of P0.05 and D-D in the 15 and 20 days of gestation group were significantly higher than those in the STBM group, and the D-D in the STBM L-NAME group was significantly shorter than that in the STBM group on the 20th day of gestation. There was no obvious change in the blank control group. In the STBM L-NAME group, he staining showed that the placental decidua was edema and inflammatory cell infiltration. There were significant differences in the expression of mRNA and protein between the basal zone group and the labyrinthine zone group. There was a significant difference in the expression of mRNA and protein between the two groups. Compared with the STBM group, the expression of sFlt-1 SEng mRNA and protein in the sFlt-1 L-NAME group was significantly higher than that in the STBM group. The proliferation and apoptosis of trophoblastic cells in the three groups were significantly different from those in the control group (P 0.05). STBM L-NAME group had the least proliferation and invasion and the most apoptosis. Compared with STBM group, the difference was statistically significant (P 0.05). Conclusion: STBM combined with L-NAME is more specific than STBM alone in constructing animal model of preeclampsia, which provides a reliable basis for clinical diagnosis and treatment of preeclampsia.
