【摘要】:目的通過生物信息學(xué)技術(shù)預(yù)測細(xì)粒棘球絳蟲Eg18抗原B、T細(xì)胞表位,為細(xì)粒棘球絳蟲篩選具有保護(hù)性抗原表位的抗原提供理論基礎(chǔ)。方法使用Expasy系統(tǒng)預(yù)測理化性質(zhì);二、三級結(jié)構(gòu)的預(yù)測使用在線預(yù)測網(wǎng)站Predict Protein;通過在線軟件ABCpred、LEPS結(jié)合二級結(jié)構(gòu)、親水性指數(shù)、表面可及性指數(shù)、柔韌性指數(shù)對B細(xì)胞表位進(jìn)行預(yù)測;應(yīng)用SYFPEITHI及IEDB軟件預(yù)測MHC-Ⅰ類HLA-A0201限制性T細(xì)胞表位。結(jié)果 Eg18抗原二級結(jié)構(gòu)中α-螺旋結(jié)構(gòu)占氨基酸殘基總數(shù)的98.1%,無規(guī)則卷曲占1.9%,無β-折疊結(jié)構(gòu);選出具有優(yōu)勢的30~41、55~71、110~126、136~144四個(gè)可能為Eg18蛋白B細(xì)胞表位的氨基酸序列區(qū)域;17~27、32~41、61~72、96~105四個(gè)較有可能形成T抗原表位的氨基酸序列區(qū)域。結(jié)論通過生物信息學(xué)預(yù)測Eg18抗原可能有4個(gè)B細(xì)胞表位且為構(gòu)象表位,有4個(gè)T抗原表位,可為進(jìn)一步了解Eg18抗原提供參考。
