【摘要】：目的探討裸鼠低位直腸癌淋巴結(jié)轉(zhuǎn)移模型血清及移植瘤組織中E-鈣黏蛋白(E-cad)的表達(dá)情況,分析其在直腸癌淋巴結(jié)轉(zhuǎn)移過程中的作用。方法取對數(shù)生長期的人大腸癌細(xì)胞株SW480接種于裸鼠直腸黏膜下,建立裸鼠低位直腸癌淋巴結(jié)轉(zhuǎn)移模型。120只裸鼠隨機(jī)分為3組,每組40只,A組(直腸癌模型組)采用二甲基肼注射+SW480種植,B組(二甲基肼對照組)采用二甲基肼注射,C組(空白對照組)不作任何處理。采用ELISA檢測血清E-cad的含量,采用Western blot法、RT-PCR法檢測種植瘤組織中E-cad蛋白及mRNA表達(dá)水平。結(jié)果A組裸鼠的血清E-Cad含量為(19.48±1.25)mg/L,明顯高于B組裸鼠和C組裸鼠的血清E-Cad含量(2.36±0.18mg/L和2.15±0.12 mg/L)(t=8.28,9.01,P0.05)。發(fā)現(xiàn)側(cè)方淋巴結(jié)轉(zhuǎn)移的裸鼠血清E-Cad含量,明顯高于未發(fā)現(xiàn)側(cè)方淋巴結(jié)轉(zhuǎn)移的裸鼠,差異有顯著性(t=10.28,P0.05)。A組裸鼠移植瘤組織中E-Cad蛋白的表達(dá)強(qiáng)度弱于B組和C組(t=9.81,7.69,P0.05)。A組中,有側(cè)方淋巴結(jié)轉(zhuǎn)移發(fā)生的裸鼠移植瘤組織中E-Cad蛋白表達(dá)強(qiáng)度較未有側(cè)方淋巴結(jié)轉(zhuǎn)移發(fā)生的裸鼠移植瘤組織中表達(dá)明顯減弱,差異有顯著性(t=9.36,P0.05)。A組裸鼠移植瘤組織中E-Cad mRNA表達(dá)明顯下調(diào),與B組和C組比較,有明顯的差異(P0.05)。A組中發(fā)現(xiàn)側(cè)方淋巴結(jié)轉(zhuǎn)移的裸鼠移植瘤組織中E-Cad mRNA表達(dá)強(qiáng)度明顯下調(diào),與未發(fā)現(xiàn)側(cè)方淋巴結(jié)轉(zhuǎn)移的裸鼠比較,差異有顯著性(t=7.85,P0.05)。結(jié)論血清E-cad的表達(dá)水平可能與直腸癌淋巴轉(zhuǎn)移密切相關(guān),同時(shí)直腸癌組織中E-Cad蛋白和mRNA表達(dá)減弱或缺失,導(dǎo)致癌細(xì)胞間粘附性降低,促進(jìn)癌細(xì)胞獲得侵襲和轉(zhuǎn)移的能力。
[Abstract]:Objective to investigate the nude mice of low rectal cancer lymph node metastasis model of E- calcium sticky protein in serum and tumor tissue (E-cad) expression and analysis of its function in the process of metastasis of rectal cancer lymph. The logarithmic growth of the human colon cancer cell line SW480 in nude mice of rectal mucosa, rectal cancer lymph node metastasis in nude mice model.120 mice were randomly divided into 3 groups, 40 rats in each group, A group (cancer model group) with two methyl hydrazine injection of +SW480 plant, B group (two methyl hydrazine group) with two methyl hydrazine injection, C group (control group) without any treatment. The content of serum E-cad ELISA, the Western blot method, RT-PCR method was used to detect the level of E-cad plant protein and mRNA expression in tumor tissue. Results the serum E-Cad content of group A in nude mice for (19.48 + 1.25) mg/L, significantly higher than that of B group and C group of nude mice in nude mice serum E-Cad content (2.36 + 0.18mg 2.15 + 0.12) /L and mg/L (t=8.28,9.01, P0.05). It is found that the lateral lymph node metastasis in nude mice serum E-Cad content was significantly higher than that found no lateral lymph node metastasis in nude mice, there was a significant difference (t=10.28, P0.05) expression of E-Cad protein in tumor tissues of nude mouse in.A group was less than that of B group and C group (t=9.81,7.69, P0.05) in the.A group, significantly decreased expression intensity is no lateral lymph node metastasis occurred in the tumor tissues E-Cad protein expression of metastasis in xenograft lateral lymph nodes, there were significant differences (t=9.36, P0.05) significantly decreased E-Cad mRNA group the expression of.A in tumor tissues of nude mouse, comparison with the B group and C group had significant difference (P0.05) intensity of E-Cad was significantly down regulated mRNA expression of lateral lymph node metastasis in nude mice transplanted tumor tissue found in the.A group, compared with lateral lymph node metastasis in nude mice and found that there were significant differences (t=7.85 P0.05). Conclusion the expression level of serum E-cad may be closely related to lymph node metastasis of rectal cancer. Meanwhile, the expression of E-Cad and mRNA in rectal cancer tissue is weakened or absent, which leads to the decrease of intercellular adhesion and the promotion of invasion and metastasis of cancer cells.
