新疆維吾爾族與哈薩克族人群17個(gè)Y-STR基因座遺傳多態(tài)性研究

【摘要】:目的:對(duì)新疆和田地區(qū)維吾爾族與阿勒泰地區(qū)哈薩克族兩民族男性Y染色體的17個(gè)Y-STR基因座的遺傳多態(tài)性進(jìn)行研究,揭示其遺傳多態(tài)性,獲得各基因座的基因頻率,了解各基因座在兩民族中個(gè)體識(shí)別率的差異,對(duì)Yfiler試劑盒在法醫(yī)學(xué)上的應(yīng)用進(jìn)行探討;同時(shí)為新疆公安機(jī)關(guān)計(jì)算和田地區(qū)維吾爾族與阿勒泰地區(qū)哈薩克族群體17個(gè)Y-STR的單倍型頻率與累積個(gè)體識(shí)別率提供基礎(chǔ)數(shù)據(jù)資料;方法:分別選取96名維吾爾族和97名哈薩克族無(wú)關(guān)男性個(gè)體血樣,Chelex法提取模板DNA,選用ABI公司AmpFISTR-YfilerTM試劑盒五色熒光復(fù)合擴(kuò)增檢驗(yàn)體系:一個(gè)反應(yīng)管中可同步擴(kuò)增l7個(gè)Y-STR基因座;并用ABI 3130XL遺傳分析儀對(duì)擴(kuò)增產(chǎn)物進(jìn)行檢測(cè)分析,使用Data Collection 3.0軟件進(jìn)行數(shù)據(jù)收集,GeneMapper ID v3.3軟件進(jìn)行分析,得到Y(jié)-STR的分型結(jié)果;結(jié)果:96例維吾爾族Y-STR基因座DYS456、DYS389Ⅰ、DYS458、DYS389Ⅱ、DYS390、DYS19、DYS385a/b、DYS392、DYS391、DYS439、DYS635、DYS393、Y-GATA-H4、DYS437、DYS438、DYS448各位點(diǎn)遺傳多樣性(GD值)分布在0.5300-0.9412之間;17個(gè)Y-STR位點(diǎn)共觀察到單倍型95種,其單倍型多樣性GD值為0.9998;97例哈薩克族Y-STR基因座各位點(diǎn)遺傳多樣性(GD值)分布在0.2769-0.8643之間;17個(gè)Y-STR位點(diǎn)共觀察到單倍型95種,其單倍型多樣性GD值為0.9996;結(jié)論:新疆和田地區(qū)維吾爾族17個(gè)Y-STR位點(diǎn)具有豐富的遺傳多樣性,選用ABI公司AmpFISTR-YfilerTM試劑盒,可為父權(quán)鑒定和父系進(jìn)化研究提供有價(jià)值的遺傳學(xué)資料;阿勒泰地區(qū)哈薩克族17個(gè)Y-STR基因座個(gè)別位點(diǎn)遺傳多樣性較差(GD值0.5),可以進(jìn)一步擴(kuò)大樣本量來(lái)進(jìn)一步驗(yàn)證YfilerTM試劑盒的17個(gè)Y-STR位點(diǎn)在該民族中的遺傳多樣性。
[Abstract]:Objective: to study the genetic polymorphism of 17 Y-STR loci in male Y chromosome of Uygur nationality and Kazak nationality in Hotan region, Xinjiang, and to reveal the genetic polymorphism and obtain the gene frequency of each locus.To understand the difference of individual recognition rate of each locus between the two nationalities, and to discuss the application of Yfiler kit in forensic medicine.At the same time, it provides basic data for the calculation of haplotype frequency and cumulative individual identification rate of 17 Y-STR of Uygur and Kazak ethnic groups in Hotan and Altay areas by Xinjiang public security organs.Methods: the template DNA was extracted by Chelex method from 96 Uygur and 97 Kazakh unrelated individual blood samples, and the five-color fluorescence multiplex amplification system of AmpFISTR-YfilerTM kit of ABI Company was used. 17 Y-STR loci could be amplified simultaneously in one reaction tube.ABI 3130XL genetic analyzer was used to detect and analyze the amplified products, and Data Collection 3.0 software was used to collect data from GeneMapper ID v3.3. The results of Y-STR typing were obtained.The GD value of haplotype diversity was 0.99989.97 Y-STR loci of Kazakh nationality were distributed in the range of 0.2769-0.8643, and 95 haplotypes were observed at 17 Y-STR loci.The GD value of haplotype diversity was 0.9996.Conclusion: 17 Y-STR loci of Uygur nationality in Hetian area of Xinjiang have abundant genetic diversity. The selection of AmpFISTR-YfilerTM kit of ABI company can provide valuable genetic data for paternity identification and paternal evolution.The genetic diversity of individual loci of 17 Y-STR loci of Kazak nationality in Altay region is poor and GD value is 0.5%, which can further expand the sample size to further verify the genetic diversity of 17 Y-STR loci of YfilerTM kit in this nationality.
