程序性細(xì)胞壞死的分子機(jī)制及其在炎癥中的作用

【摘要】:越來(lái)越多的研究表明,細(xì)胞壞死是一種受到精密調(diào)控的"新型"程序性細(xì)胞死亡方式。當(dāng)細(xì)胞凋亡不能正常發(fā)生而細(xì)胞必須死亡時(shí),壞死作為凋亡的"替補(bǔ)"方式被激活。程序性細(xì)胞壞死主要由腫瘤壞死因子受體(tumor necrosis factor receptor,TNFR)家族以及Toll樣受體(Toll-like receptor,TLR)家族啟動(dòng),并通過(guò)和受體蛋白互作的兩個(gè)蛋白激酶RIP1(receptor interacting protein kinase 1)和RIP3(receptor interacting protein kinase 3)傳遞死亡信號(hào),募集并磷酸化MLKL(mixed lineage kinase domain-like protein),而MLKL作為細(xì)胞死亡的執(zhí)行者最終會(huì)導(dǎo)致壞死的發(fā)生。壞死的細(xì)胞會(huì)向周?chē)尫牌鋬?nèi)容物,這些內(nèi)容物作為DAMPs(damage-associated molecular pattern molecules)可刺激周?chē)?xì)胞發(fā)生炎癥反應(yīng),激活機(jī)體免疫應(yīng)答。該文以TNF-α誘導(dǎo)的細(xì)胞壞死途徑為出發(fā)點(diǎn),著重闡述程序性細(xì)胞壞死的分子機(jī)制及其在炎癥中的作用,并回顧和展望了其在臨床診療中的可能性。
[Abstract]:More and more studies show that cell necrosis is a kind of "new" by the precise regulation of programmed cell death. Apoptosis can occur when normal cells must be activated as death, necrosis and apoptosis of "substitute". Programmed cell necrosis mainly by tumor necrosis factor receptor (tumor necrosis factor receptor. TNFR) family and Toll like receptor (Toll-like receptor TLR) family started, two protein kinase RIP1 and the receptor protein interaction (receptor interacting protein kinase 1) and RIP3 (receptor interacting protein kinase 3) transmit death signal, recruitment and phosphorylation of MLKL (mixed lineage kinase domain-like protein), and MLKL as the executor of cell death eventually lead to necrosis. Necrosis of the cells will release its contents into the surrounding, the content of DAMPs (damage-associated as mole Cular pattern molecules) can stimulate the peripheral cell inflammation, activate the immune response. In this paper, way of cell necrosis induced by TNF- as a starting point, focuses on the molecular mechanisms of programmed cell necrosis and its role in inflammation, and discussed its possibility in the clinical diagnosis and treatment and review.
