【摘要】：目的探討β3腎上腺受體(β3-adrenoceptor,p3-AR)在人子宮平滑肌中的表達及與分娩發(fā)動的關(guān)系。 方法選擇2011年6月～2012年2月在中南大學湘雅二醫(yī)院住院妊娠足月未臨產(chǎn)產(chǎn)婦15例(未臨產(chǎn)組,not in labor,NL組)、足月臨產(chǎn)產(chǎn)婦15例(臨產(chǎn)組,in labor,L組)、未妊娠因?qū)m頸上皮內(nèi)瘤樣變行子宮全切術(shù)婦女15例(未妊娠組,in non-pregnant,NP組)。應(yīng)用實時熒光定量RT-PCR和免疫印跡(Western Blot)檢測3組婦女子宮平滑肌中p3-AR的mRNA和蛋白的表達水平。 結(jié)果 1、妊娠足月未臨產(chǎn)組、足月臨產(chǎn)組、未妊娠組均有p3-AR的表達； 2、足月臨產(chǎn)組3-AR mRNA的相對表達量(0.317+0.06)明顯低于足月未臨產(chǎn)組(0．714±0.103),P=0.000,P0.05。足月未臨產(chǎn)組表達明顯高于未妊娠組(0.282+0.075),P=0.000,P0.05。非妊娠組與足月臨產(chǎn)組相比無顯著差異,P=-0.25,P0.05。 3、足月臨產(chǎn)組D3-AR蛋白的相對表達量(0.379±0.027)明顯低于足月未臨產(chǎn)組(0.690±0.054),P=0.000,P0.05。足月未臨產(chǎn)組蛋白相對表達量明顯高于未妊娠組(0.362+0.045),P=0.000,P0.05。未妊娠組與足月臨產(chǎn)組相比無顯著差異,P=0.3,P0.05。 結(jié)論1、子宮平滑肌中有p3-AR的表達；2、子宮平滑肌中p3-AR表達減少與分娩發(fā)動有關(guān)。
[Abstract]:Objective to investigate the expression of 尾 3 adrenoceptor p3 ARR in human uterine smooth muscle and its relationship with labor initiation.Methods from June 2011 to February 2012, 15 pregnant women who were hospitalized in Xiangya No.2 Hospital of Central South University (not in labor group) and 15 women in term delivery group (labor group) were selected.Total hysterectomy was performed in 15 women with tumor-like degeneration (non-pregnant NP group).The expression of p3-AR mRNA and protein in the uterine smooth muscle was detected by real-time fluorescence quantitative RT-PCR and Western blotting.Result1. The expression of p3-AR was found in the full-term labor group, the term labor group and the non-pregnancy group.2, the relative expression of 3-AR mRNA in term labor group (0.317 0.06) was significantly lower than that in term non-labor group (0.714 鹵0.103).The expression of P0. 282 0. 075 was significantly higher in term non-parturient group than that in non-pregnant group (P 0. 000 P 0. 05).There was no significant difference between non-pregnancy group and term labor group.The relative expression of D3-AR protein in term labor group (0.379 鹵0.027) was significantly lower than that in non-term labor group (0.690 鹵0.054).The relative expression of protein in term unpartum group was significantly higher than that in unpregnant group 0.362 0.045% P0. 000 P0. 05.There was no significant difference between non-pregnancy group and term labor group.Conclusion 1. There is p3-AR expression in uterine smooth muscle. The decrease of p3-AR expression in uterine smooth muscle is related to the onset of labor.
