RNF34調(diào)控天然免疫的初步研究

【摘要】:目的研究環(huán)指蛋白34(RING finger protein 34,RNF34)對(duì)天然免疫的調(diào)控。方法利用重組PCR方法構(gòu)建pcDNA3-Flag-RNF34、pcDNA3-Flag-RNF34-ΔFYVE、pcDNA3-Flag-RNF34-ΔCID和pcDNA3-Flag-RNF34-ΔRING質(zhì)粒,并在HEK293T細(xì)胞中瞬時(shí)表達(dá);利用雙熒光素酶報(bào)告基因技術(shù)檢測(cè)RNF34及其3種突變體對(duì)仙臺(tái)病毒(SeV)和N-RIG-Ⅰ激活NF-κB和IFN-β轉(zhuǎn)錄活性的影響。結(jié)果成功構(gòu)建了Flag-RNF34及其3種結(jié)構(gòu)域缺失突變體的真核表達(dá)質(zhì)粒;發(fā)現(xiàn)在SeV刺激下,RNF34對(duì)NF-κB和IFN-β轉(zhuǎn)錄活性有明顯抑制作用,RNF34-ΔFYVE、RNF34-ΔCID和RNF34-ΔRING與RNF34相比此種抑制作用減弱。同時(shí)發(fā)現(xiàn)對(duì)于N-RIG-Ⅰ激活的NF-κB和IFN-β活性,RNF34及其3種結(jié)構(gòu)域缺失突變體也有相似的抑制作用。結(jié)論 RNF34通過負(fù)調(diào)控RIG-Ⅰ-MAVS信號(hào)通路調(diào)控天然免疫。
