【摘要】：目的探討Toll樣受體5(TLR5)對間充質(zhì)干細胞(MSC)生物學特性的影響,以期探究TLR5在MSC與腫瘤關(guān)系中的作用。方法構(gòu)建過表達TLR5的慢病毒載體并用相關(guān)病毒感染MSC,通過熒光表達量、RT-q PCR和Western印跡驗證TLR5在MSC中的過表達情況,通過MSC-TLR5增殖、表面抗原和細胞分化來驗證TLR5過表達對MSC生物學性狀影響,通過檢測CBLB502激活TLR5后白細胞介素6(IL-6)和IL-8的表達水平驗證TLR5生物學功能。結(jié)果 TLR5慢病毒感染MSC后,mRNA水平和蛋白水平檢測TLR5表達量均顯著升高,MSC-TLR5增殖、免疫表型和分化能力未發(fā)生改變,同時,CBLB502激活TLR5后,IL-6和IL-8表達量顯著升高(P0.001)。結(jié)論攜帶TLR5的慢病毒感染MSC后不影響MSC生物學功能,TLR5具有功能活性且可激活下游信號通路發(fā)揮免疫調(diào)節(jié)作用。
[Abstract]:Objective to investigate the effects of Toll like receptor 5 (TLR5) on the biological characteristics of mesenchymal stem cells (MSCs). Methods Lentivirus vector expressing TLR5 was constructed and infected with TLR5. The overexpression of TLR5 in MSC was verified by fluorescent expression of RT-q PCR and Western blotting, and proliferation of TLR5 by MSC-TLR5. Surface antigen and cell differentiation were used to verify the effect of TLR5 overexpression on the biological characteristics of MSC. The biological function of TLR5 was verified by detecting the expression level of interleukin-6 (IL-6) and IL-8 after TLR5 activated by CBLB502. Results the expression of TLR5 in MSC infected with TLR5 lentivirus significantly increased the proliferation of MSC-TLR5. The immunophenotype and differentiation ability did not change. At the same time, the expression of IL-6 and IL-8 increased significantly after activation of TLR5 by CBLB502. Conclusion Lentivirus carrying TLR5 does not affect the biological function of MSC after MSC infection and can activate downstream signaling pathway to play an immunomodulatory role.
