【摘要】:目的研發(fā)一種基于環(huán)介導(dǎo)等溫?cái)U(kuò)增(1oop-mediated isothermal amplification,LAMP)技術(shù)原理的組織胞漿菌感染的分子診斷技術(shù)。方法以組織胞漿菌為研究對(duì)象,針對(duì)其種特異性的M抗原基因序列,設(shè)計(jì)并篩選數(shù)套基于LAMP技術(shù)的特異性引物,同時(shí)優(yōu)化其反應(yīng)條件,建立一種針對(duì)組織胞漿菌感染的快速分子鑒定技術(shù)。結(jié)果該LAMP技術(shù)在65℃、90min反應(yīng)條件下,對(duì)組織胞漿菌基因組DNA的檢測(cè)敏感度達(dá)到5.3pg/反應(yīng)(即5.3×10-12 g,約160個(gè)基因組DNA拷貝),且與其相近種屬真菌無交叉反應(yīng)。結(jié)論我們研發(fā)的這種基于LAMP技術(shù)原理的組織胞漿菌分子診斷技術(shù)具有操作簡(jiǎn)便、特異性及敏感性高等優(yōu)點(diǎn),具有臨床推廣應(yīng)用的潛力。
