游泳對高脂膳食誘導(dǎo)肥胖和肥胖抵抗模型大鼠的作用

【摘要】:背景:高脂膳食誘導(dǎo)的肥胖動(dòng)物是研究人類肥胖的首選模型,瘦素及其受體的表達(dá)在高脂膳食誘導(dǎo)的肥胖和肥胖抵抗中發(fā)揮重要作用,但其確切機(jī)制和游泳對它的作用還不清楚。目的:在肥胖和肥胖抵抗的基礎(chǔ)上,探討7周游泳運(yùn)動(dòng)對高脂膳食大鼠血清瘦素及下丘腦瘦素受體表達(dá)的影響。方法:60只SD大鼠高脂飼料喂養(yǎng)8周,根據(jù)體質(zhì)量篩選出肥胖和肥胖抵抗大鼠。取14只肥胖大鼠等分為肥胖組和肥胖運(yùn)動(dòng)組,取14只肥胖抵抗大鼠等分為肥胖抵抗組和肥胖抵抗運(yùn)動(dòng)組。各組大鼠繼續(xù)高脂膳食喂養(yǎng)。肥胖運(yùn)動(dòng)組和肥胖抵抗運(yùn)動(dòng)組大鼠同時(shí)進(jìn)行連續(xù)7周的自由游泳運(yùn)動(dòng)訓(xùn)練。結(jié)果與結(jié)論:與肥胖組相比,肥胖運(yùn)動(dòng)組大鼠體質(zhì)量、脂肪量、脂體比、血清瘦素濃度顯著降低,下丘腦瘦素受體的基因表達(dá)水平顯著升高;而與肥胖抵抗組相比,肥胖抵抗運(yùn)動(dòng)組大鼠上述指標(biāo)無明顯變化。說明游泳能夠增加肥胖大鼠機(jī)體的能量消耗,加快脂肪分解,降低血液中瘦素濃度,有效增加下丘腦瘦素受體的基因表達(dá)水平,從而緩解瘦素抵抗,改善機(jī)體代謝水平。
[Abstract]:Background: obese animals induced by high fat diet are the first choice to study human obesity. The expression of leptin and its receptor plays an important role in obesity and obesity resistance induced by high fat diet. But the exact mechanisms and effects of swimming on it are unclear. Objective: based on obesity and obesity resistance, To investigate the effects of swimming for 7 weeks on the expression of leptin in serum and leptin receptor in hypothalamus of rats with high fat diet. Methods Sixty SD rats were fed with high fat diet for 8 weeks. Obesity and obesity resistant rats were selected according to body mass. Fourteen obese rats were divided into obese group and obese exercise group. 14 obese resistant rats were divided into obesity resistance group and obesity resistance exercise group. Rats in each group were fed with high fat diet. Rats in obese exercise group and obesity resistance exercise group were trained in free swimming for 7 weeks at the same time. Results and conclusions: compared with the obese group, In obese exercise group, the body weight, fat content, fat body ratio, serum leptin concentration and the expression level of leptin receptor gene in hypothalamus were significantly decreased, but compared with those in obese resistance group, the expression of leptin receptor gene in hypothalamus was significantly increased in obese exercise group. The results showed that swimming could increase the energy consumption, accelerate fat decomposition, decrease the concentration of leptin in blood and increase the gene expression level of leptin receptor in hypothalamus of obese rats, which indicated that swimming could increase the energy consumption, accelerate fat decomposition, decrease the concentration of leptin in blood, and increase the expression of leptin receptor gene in hypothalamus. So as to alleviate leptin resistance, improve the metabolism level of the body.
